柑橘属果实中类黄酮成分的高效液相色谱标准化测定方法

Determining Flavonoids in Citrus Fruits Using HPLC Standardized Method

摘要: 针对目前尚无柑橘属果实中黄酮类化合物标准化测定方法,建立了高效液相色谱同时测定柑橘属果实中类黄酮成分柚皮苷、橙皮苷和新橙皮苷含量的标准化方法。以Waters Atlantis T3- C18色谱柱 (4.6×150 mm,3μm)分离,柱温40℃;以乙腈-0.5%乙酸溶液(体积比为20:80)为流动相,流速为1.0 mL/min;采用二极管阵列检测器,检测波长283nm。柚皮苷、橙皮苷和新橙皮苷的线性范围分别为1~500 μg/mL(r=0.9999),1~500 μg/mL ( r=1.0000),1~500 μg/mL (r=0.999 9),加标回收率都在95%以上。方法学考察结果显示符合测定要求,并适用于柑橘属果实的测定。该方法操作简便、快速,结果可靠,重现性好,可用来研究柑橘属果实中的类黄酮成分含量。

Abstract: For determining narigin, hesperidin and neohesperidin in Citrus fruits, a high performance liquid chromatographic (HPLC) method was established. The HPLC condition was as follows: Waters Atlantis T3-C18 column (4.6 mm×150 mm, 3 μm) was used at 40℃ with acetonitrile-water containing 0.5% acetic acid (20:80, v/v) as the mobile phase with a flow rate of 1.0 mL/min, diode array detector with the detection at 283 nm. The linear range of naringin, hesperidin and neohesperidin was 1- 500 μg/mL (r=0.9999), 1- 500 μg/mL (r=1.0000), 1-500 μg/mL (r=0.9999), respectively. The average recovery was all over 95%. The results of methodological study demonstrate that the method could meet the determination requirements, and is convenient and accurate with good reproducibility, could be used to study the content of flavonoids in Citrus fruits.
