高抗寒毛白杨杂种离体培养及玻璃化苗研究

Populus tomentosa Hybrid with High Cold Resistance: In vitro Culture and Vitrification Seedlings Research

摘要: [目的]为了能够将毛白杨优良资源应用到北纬41°以北地区,为我国北方生态防护林建设提供优良苗木,特进行高抗寒性毛白杨杂种—‘蒙树1号杨’组织培养技术研究。[方法]利用‘蒙树1号杨’切枝水培嫩芽为外植体,开展了最佳消毒剂和消毒时间筛选,以及无菌苗茎段增殖、叶片分化、生根培养和玻璃化苗恢复培养研究。[结果]嫩芽在0.10%升汞中灭菌5 min效果最佳,外植体污染率为15.60%、萌芽率达77.70%;最佳茎段增殖培养基为MS + 0.20 mg/L 6-BA + 0.05 mg/L NAA,平均分化出芽4.21个;不定根最佳诱导培养基为1/2 MS + 0.50 mg/L IBA + 0.20 mg/L NAA,生根率达93.90%;玻璃化苗在MS + 0.2 mg/L 6-BA + 0.05 mg/L NAA + 30 g/L蔗糖+8 g/L琼脂培养基中恢复效果最好,恢复率达82.38%。[结论]‘蒙树1号杨’离体快繁体系的建立,为毛白杨体细胞多倍体育种及我国西北地区园林绿化和生态建设夯实了基础。

Abstract: To apply the good resources of Populus tomentosa in the north of north latitude 41°, and provide high quality seedlings for the construction of ecological protection forest in northern China, the shoots from branch water culture of‘Mengshu No.1’(P. tomentosa hybrid with high cold resistance) were used as explants, the best disinfectant and sterilization time were screened. The stem segments propagation of aseptic seedlings, leaf blade differentiation, rooting culture and vitrified seedling culture were conducted. The shoots were sterilized in 0.10% HgCl for 5 minutes, the disinfectant effect was the best. The pollution rate of explants was 15.60%, and the germination rate was 77.70%. The best proliferation medium of stem segments was MS+ 0.20 mg/L 6-BA+0.05 mg/L NAA, and the average number of differentiation bud was 4.21. The best induction medium of root was 1/2 MS+0.50 mg/L IBA+0.20 mg/L NAA, and the rooting rate was 93.90%. Vitrification seedlings restored best in the medium MS+0.2 mg/L 6-BA + 0.05 mg/L NAA + 30 g/L sucrose+8 g/L AGAR, and the restoration rate was 82.38%. The establishment of in vitro micropropagation system for‘Mengshu No.1’ lays a foundation of somatic polyploid breeding for P. tomentosa, landscaping and ecological construction in northwest China
