小金海棠超氧化物歧化酶基因MxSod2克隆与序列分析

摘要: 本实验以苹果属植物小金海棠(Malus xiaojinensis Cheng et Jiang)为试材,利用差异筛选消减cDNA文库和RACE相结合的方法,分离到了小金海棠超氧化物歧化酶基因MxSod2(Genbank登录号为 DQ431473),该基因全长805个碱基,开放阅读框为471bp,编码157个氨基酸,推测分子量15kDa。该基因具有CuZn-SOD基因的典型结构域特征。通过系统进化树分析,小金海棠MxSod2与龙眼、荔枝等非禾本科植物保持了较近的亲缘关系,与花生、水稻、玉米等禾本科植物亲缘关系较远。

Abstract: A superoxide dismutase gene was cloned by differential screening of the subtracted cDNA library and RACE from Malus xiaojinensis Cheng et Jiang, which was named MxSod2 (Accession No. DQ431473). The full-length cDNA of MxSod2 is 805 bp, including the open reading frame (ORF) of 471 bp. It encodes a protein of 157 amino acids, with a molecular weight of 15 kDa. The MxSod2 contained the conserved domains shared in all known SOD. The phylogenetic tree suggested that MxSod2 was closer to the SOD of non-graminaceous plants including longan and litchi, but farther with the SOD of peanut, rice and maize.
