Mature Embryo Culture in Waxy Maize

摘要: 以糯玉米成熟胚为外植体作离体培养,研究在培养基中添加植物激素,硝酸银及有机物等对玉米愈伤组织的诱导和胚性愈伤继代生长的影响。结果表明:在诱导培养阶段,N6+6-BA 0.2mg/L+2,4-D 2mg/L的培养基诱导率最高,愈伤质量好;在继代培养阶段,MS+6-BA 0.2mg/L+2,4-D 2mg/L +AgNO310mg/L或CW (Coconut Water) 10%的培养基可改善胚性愈伤的质量。

Abstract: Mature waxy maize embryos were cultured as explants in vitro, and the effects on callus induction and subculturing were investigated under culture mediums with different additives, such as plant hormones, AgNO3, organic compounds, etc. The results showed that the combination of N6 + 0.2mg/L 6-BA + 2mg/L 2,4-D could obtain the most calli of higher quality in the induction phase, while the combination of MS + 0.2mg/L 6-BA + 2mg/L 2,4-D +10mg/L AgNO3 or 10% CW could make sure the calli of higher quality further in the stage of subculturing.
