双黄连对犬肝细胞对乙酰氨基酚损伤的保护作用

Protective effect of Shuanghuanglian on acetaminophen injuried Canine hepatocytes

摘要:

胶原酶两步灌流法分离犬的肝细胞,对乙酰氨基酚(AP)制备犬肝细胞损伤模型,不同浓度的双黄连处理,测定培养上清液中ALT和AST的活性。Western blot法测定犬肝细胞中细胞色素酶 P450的表达水平。结果显示:16mM AP作用12h可引起肝细胞内ALT和AST的含量显著升高,说明16mM AP是导致犬肝细胞损伤的最适浓度。1-5mM双黄连处理损伤的犬肝细胞,ALT和AST的活性显著下降。处理48h时,CYP450的表达随双黄连浓度的增加而升高,1-5mM时表达最佳。说明1-5mM双黄连具有很好的保护对乙酰氨基酚所致的犬肝细胞损伤的作用。

Abstract:

Canine primary cultured hepatocytes were isolated by a modified two-step collagenase digestion method. The injuried hepatocyte model was made by acetaminophen. The injuried hepatocytes were treated by Shuanghuanglian, ALT and AST activities were measured, the expression of CYP450 was determined by Western blot. The results as follows, the activities of ALT and AST in increased significantly by 16mM AP treated for 12h. The activities of ALT and AST decreased significantly. The expression of cytochrome P450 in injuried Canine hepatocytes increased gradually for 48h, the best dose was 1-5mM. 1-5mM Shuanghuanglian had protective effect on acetaminophen injuried Canine hepatocytes.
