摘要：

为了成功建立牛滋养层干细胞样细胞系，探讨牛滋养层干细胞样细胞（Bovine Trophoblastic Stem-like Cells）体外培养条件及相关影响因素，以胎牛子宫成纤维细胞条件培养液作为培养基，胶原铺被培养皿，利用体外受精牛囊胚培养得到牛滋养层干细胞样细胞，并调整培养体系，以探讨其最适生长条件。结果表明：利用添加FBS、β-巯基乙醇、肝素以及胎牛子宫成纤维细胞条件培养液的DMEM/F12，体外培养牛囊胚，可以初步建立牛滋养层干细胞样细胞系。

Abstract:

The study aimed to investigate the proper conditions and influential factors in order to best establish a trophoblastic stem-like cell line, derived from in vitro matured and fertilized blastocysts. The cells were cultured on collagen coated wells using fetal bovine fibroblast-conditioned medium with an adjustable culture system used to investigate the effect on cell growth. We found the the DMEM/F12 medium was mixed with fetal bovine fibroblast-conditioned medium at a dilution of 1:1 and supplemented with FBS, Heparin and β-mercaptoethanol was the best culture medium.
