A Kind of Simple and Efficient DNA Recovery Method from Polyacrylamide Gel

本文介绍了一种简单的聚丙烯酰胺凝胶DNA回收方法(直取法),通过与传统煮沸法比较以证明直取法回收聚丙烯酰胺凝胶DNA的高效性。以木薯cDNA为材料,将直取法与常规煮沸法从实验方法、杂带污染率以及回收扩增率三个方面进行比较。实验结果表明:直取法较煮沸法简单、快速;煮沸法的杂带污染率为59.09%,直取法为43.08%;煮沸法的平均产物量约为40 ng/μL,直取法约为60 ng/μL。研究结果表明,直取法相对煮沸法简单,快速,扩增效率高,杂带污染几率小。

Abstract:

This article described a simple recovery method of DNA from polyacrylamide gels, direct recovery method. The aim was to prove the high efficiency of direct recovery method for DNA recovery from polyacrylamide gels, compared with traditional boiling method. With cassava cDNA as experimental material, this method and boiling method were compared from three aspects: experimental method, extract and amplified rates, pollution rates. The results showed that direct recovery method was more simple and fast than boiling method, pollution rates of boiling method was 59.09%, and the direct recovery method was 43.08%, the average amplification products by boiling method about 40 ng/μL, the direct recovery method was 60 ng/μL. The results indicated that the direct recovery method was easier, faster, and efficient. It had lower pollution rates.
