鼓槌石斛组织培养及快繁技术研究

Research on the Tissue Culture and Rapid Propagation Technology of Dendrobiun chrysotoxum Lindl

摘要: 以鼓槌石斛成熟果实中的种子为外植体,研究了鼓槌石斛无菌苗增殖,壮苗生根的最佳培养基配方,并对无菌苗增殖培养条件进行了优化。研究结果表明:可通过种子直接形成无菌苗、再经无菌苗增殖、壮苗生根的途径,生成完整植株,实现鼓槌石斛种苗的规模化生产;并筛选出适宜的培养基:无菌苗增殖培养基为MS+0.5mg/LNAA+2.0mg/LBA+50g/L香蕉泥,增殖系数高达4.7,且长势良好、芽苗健壮;壮苗生根培养基为1/2MS+0.3mg/LNAA,生根苗根系长而粗壮,叶色浓绿。无菌苗增殖的优化条件:白糖浓度为30g/L,有机添加物为香蕉泥,丛植芽数量为5株/丛时增殖系数最高,幼苗长势最好。

Abstract: he seeds from mature fruit of Dendrobiun chrysotoxum Lindl were used as explants to study the suitable medium for aseptic seedlings proliferation and rooting, and the culture condition of aseptic seedlings proliferation was optimized. The results showed that aseptic seedling could be formed directly from seeds, then through its proliferation and rooting to generate complete plants, thus to realize the large-scale production of Dendrobiun chrysotoxum seedling. The suitable culture medium was selected, the optimal medium for aseptic seedling proliferation was MS+0.5 mg/L NAA+2.0 mg/L BA+50 g/L banana puree, proliferation coefficient was 4.7 and they grew well; while the optimal medium for rooting was 1/2MS+0.3 mg/L NAA, the roots of root seedlings were long and thick, the leaves were also thick. Optimization conditions of proliferation: the proliferation coefficient was the highest and plants grew the best while the suitable sugar concentration was 30 g/L, the suitable organic additive was banana puree and the suitable clump planting bud number was 5 each.
