Determination of the Isomers of Vitamin E by Normal Phase High Performance Liquid Chromatography

摘要: 采用正相高效液相色谱法同时分离和测定生育酚和生育三烯酚的含量,确定一种简单准确测定维生素E异构体含量的方法。样品经前处理后,用Zorbax NH2柱分离,以正己烷-异丙醇为流动相洗脱,在波长292 nm处进行检测,采用外标法对七种维生素E异构体:α-生育酚、α-生育三烯酚、β-生育酚、γ-生育酚、γ-生育三烯酚、δ-生育酚和δ-生育三烯酚进行了定量分析。结果表明,各组分线性关系良好(相关系数0.9993~0.9998),精密度高,稳定性和重现性好,加样回收率在97.0%~102.1%,方法简便准确,可用于不同样品的定量分析。

Abstract: Tocopherols and tocotrienols were separated and determined by NP-HPLC to establish a simple and accurate method to determine the contents of the vitamin E isomers. The sample was purified and then separated on the Zorbax NH2 column. The mobile phase consisted of hexane and isopropanol. The detection was at 292nm. Seven isomers of vitamin E:α-tocopherol、α-tocotrienol、β-tocopherol、γ-tocopherol、γ-tocotrienol、δ-tocopherol and δ-tocotrienol were identified and calculated. The results showed that the corelation coefficients were from 0.9993 to 0.9998. The method suggested the high degree of precision, stability and the repeatability, and the recoveries were from 97.0% to 102.1%. This method was simple, accurate and suitable for the analysis of vitamin E in the whole berry oil of sea buckthorn.
