蒿属药用植物叶绿体上的psbA-trnH序列分析

摘要: 【摘要】 目的:采用叶绿体上psbA-trnH序列中的信息,运用物种特异的DNA序列信息位点区分黄花蒿和常见同属植物的混淆种茵陈蒿、青蒿,为药用植物的鉴定提供科学依据。方法:提取被测植物的DNA,使用PCR方法扩增psbA-trnH序列片段,测定并比较所得序列,软件分析得到上述三种植物各自特有的DNA序列特征位点。结果:黄花蒿和茵陈蒿、青蒿植物之间psbA-trnH序列虽具有较高的相似性,但也有显著的特征性的不同,可以依靠它们各自特有的信息位点区分开来。结论:psbA-trnH序列中的信息位点可以用来区分黄花蒿、茵陈蒿和青蒿植物。该方法首次为蒿属药用植物的分子鉴定提供了可靠、简便的参考依据。

Abstract: [Abstract] Objective:This research focused on analyzing the differences of psbA-trnH region sequences of Artemisia annua L,Artemisia capillaris Thunb and Artemisia carvifolia Buch, which can provide the species-special character-sites for discriminating them. Method:DNA was extracted from the collected three Artemisia samples and their psbA-trnH regions were amplified by PCR,and then sequenced and analyzed to find the character-sites.Result:Although the psbA-trnH regions among the three species are high similar, there are several character-sites for themselves, which can be used in discriminating them. Conclusion:psbA-trnH region can provide the character-sites used to identify each of the three varieties unequivocally. It is the first time to use DNA technology in drug species’ discrimination of Artemisia, which provided a reliable and simple reference for the authentication of them.
