海南沼虾ITS-1序列分析

The Analysis of ITS-1 Sequence for Macrobrachium Hainanense

摘要: 为了了解海南沼虾遗传方面的信息,本文利用ITS-1分子标记技术对珠江海南沼虾群体作了研究。研究结果表明:①其ITS-1片段在1700bp~1800bp之间,较日本沼虾短;②其碱基含量变化范围为:27%~30%(A)、27%~31%(G)、14~16%(C)、43%~46%( G+ C);③在海南沼虾ITS-1序列中,包含着非常丰富的SSR序列信息,具体为(AG)n、(GA)n、(AGT)n、(AGC)n、(GCA)n等重复,在个体中出现的比例分别为100%、100%、25%、12.5%、12.5%。同时(AG)n、(GA)n重复出现了较高多态性; ④BLAST比对结果表明,海南沼虾同日本沼虾和刀额新对虾都有较高的同源性。本研究结果在一定程度上填补了海南沼虾ITS-1方面研究的空白,同时也为海南沼虾后续研究提供了理论参考。

Abstract: In order to obtain genetic information of Macrobrachium Hainanense, this paper focus on the research by ITS-1 molecular marker technology. As showed by results: ①The length of ITS-1 is range from 1700bp to 1800bp, shorter than Macrobrachium nipponense.②The contents scope of bases(A,G, C ,G+ C) is 27%~30%,27%~31%,14~16%,43%~46% respectively. ③ Rich SSR information is contained in ITS-1 sequence for Macrobrachium Hainanense, such as repeats of (AG)n, (GA)n, (AGT)n, (AGC)n, (GCA)n and repeats of (AG)n, (GA)n are polymorphism locus. The proportion of which appear among individuals are 100%,100%,25%,12.5%,12.5%. ④Homology is demonstrated by BLAST between Macrobrachium Hainanense and Macrobrachium nipponense, Metapenaeus ensis. This article is that not only fills up Macrobrachium Hainan ITS-1 research gaps, but also provides a theoretical reference for Macrobrachium Hainan follow-up studies.
