利用荧光定量PCR方法研究猪繁殖与呼吸综合征病毒在Marc-145细胞的增殖规律

Study on the replication of Porcine Reproductive and Respiratory Syndrome Virus in Marc-145 Cells using Fluorescence Quantitative PCR

摘要:猪繁殖与呼吸综合征病毒可以在原代猪肺泡巨噬细胞(PAM)、CL2621细胞系及Marc-145细胞系上生长繁殖,但对于其增殖规律不甚了解。试验利用实时荧光定量PCR技术,研究猪繁殖与呼吸综合征病毒在Marc-145细胞中的增殖规律;结果发现:病毒在接种后12 h开始大量增殖,在18-36 h增殖最为迅速,在36-48 h病毒增殖达到最高峰;研究还发现在18 h 时Marc-145细胞开始释放病毒,48 h达最高峰。该结果为研究猪繁殖与呼吸综合征病毒的致病机理及利用细胞毒生产疫苗提供试验基础。

Abstract:

Abstract: Porcine Reproductive and Respiratory Syndrome Virus (PRRSV) could grow in PAM, the cell line of CL2621 and Marc-145, but CPE is not distinctness, and the replication disciplinarian is still clearly unknown. This experiment investigated that the replication disciplinarian of PRRSV in the line of Marc-145 using Fluorescence Quantitative PCR. The results found that the virus begin to replicate in a great speed at the 12 h after inoculation and its growth is fastest between the 18-36 h, reaches peak at 48 h. These experiments supplied the production of vaccine and helps to do research on viral pathology of PRRSV.
