多基因表达载体KCTB转化宁夏枸杞的研究

摘要:

为研究多基因表达载体KCTB转化宁夏枸杞,本研究利用农杆菌介导技术,利用叶盘转化法,将构建有GUS、BAR、KC1、CAF14个功能基因的表达载体KCTB转化宁夏枸杞,并获得了23株抗性植株;GUS组织化学染色显示,在转基因植株的根、茎、叶中都有表达,PCR结果显示,分别检测到了BAR、KC1、CAF1基因,说明多基因表达载体KCTB已经完全整合到了宁夏枸杞的基因组中。

Abstract:

For researching transformation of Lycium barbarum. L ,the multigene expressed vector KCTB that constructed with four functional genes, GUS、BAR、KC1、CAF1 was introduced into Lycium barbarum. L mediated with agrobacterium tumefaciens. Analysis of transgenic plant by histochemical staining showed that the GUS was expressed predominately in leaves, and also lesser degree in roots. The result of PCR suggest that BAR、KC1、CAF1 was detected and also the KCTB was contained in genome of Lycium barbarum. L.
