摘要:

采用CTAB、SDS、改良CTAB法对五味子总DNA提取进行研究,并对总DNA进行电泳分析和含量测定,结果表明以改良CTAB法提取五味子叶片DNA纯度最好,以此DNA为模板可获得条带清晰、重复性好的RAPD扩增结果。从14条重复性好的引物中共扩增出67条带,平均每个引物4.8个片段,多态性位点数为58,比例为86.6%。UPGMA聚类分析的结果与五味子形态学分类结果相似,10份五味子优系在遗传相似系数0.69处可划分为3个类群,该RAPD技术体系可以很好地用于10份五味子种质资源的鉴定。

Abstract:

Genomic DNA of Schisandra chinensis (Turcz.) Baill. was extracted by means of three different DNA extraction methods, CTAB, SDS and improved CTAB methods. The purity and quality of DNA obtained by these methods were compared, and the quality was evaluated by agarose gel electrophoresis. The results showed that DNA sample obtained by improved CTAB method had the highest purity and quality. Good repeatability of RAPD amplification can be obtained with this DNA template. Fourteen random primers of good repeatability generated 67 bands, with an average of 4.8 fragments per primer, and 86.6% of them were polymorphic. UPGMA analysis showed similar results with the morphological classification. The accessions can be divided into three groups at the coefficient 0.69. The RAPD technology system can be used to identify 10 Schisandra chinensis (Turcz.) Baill. accessions.
