人工栽培石斛的ISSR标记分析

ISSR Analysis of Cultivated Medicinal Species of Dendrobium Sw.

摘要:

为了给石斛品种的人工选育和杂交育种提供参考,利用ISSR分子标记技术对9种药用栽培石斛的遗传多样性进行了研究。从备选的引物中筛选出14条多态性引物,共检测到179个多态性位点,多态性比率为94.71%。Jaccard相似性指数显示:15个铁皮石斛样本的相似度介于0.264~0.536之间,6个齿瓣石斛样本的相似度介于0.351~0.561之间,不同种间的相似度介于0.115~0.371之间。虽然种内与种间的相似度值存在一些交叉情况,但UPGMA聚类分析表明,不同种的石斛之间并不存在明显的杂交情况。ISSR标记检测结果显示,源于不同栽培地区栽培的的石斛属植物的遗传多样性非常丰富。

Abstract:

To provide the reference for variety selection and cross breeding, the genetic diversity of Dendrobium was analyzed by ISSR markers. Fourteen ISSR primers were screened for the ISSR-PCR reaction, 179 polymorphic loci were detected, and the polymorphic percentage was 94.71%. Jaccard similarity index showed that the similarity index among D. candidum was 0.264-0.536, 0.351-0.561 among D. devonianum and 0.115-0.371 among different species. The result of UPGMA analysis showed it had no obvious cross breeding between intraspecies and interspecies. The ISSR results indicated the genetic diversity of Dendrobium plants from different areas was rich.
