基于细胞色素B(Cyt b)基因对翘嘴鲌不同群体遗传多样性的分析

Culter alburnus Basilewsky in Different Populations: Genetic Diversity Analysis Based on Cytochrome B (Cyt b) Gene

摘要:

为了解长江下游翘嘴鲌(Culter alburnus Basilewsky)不同群体遗传多样性水平,评估其资源状况。研究以淀山湖(DSH)、太湖(TH)、长荡湖(CD)和长江(CJ)4个水域共211尾翘嘴鲌为研究对象,对其线粒体DNA细胞色素b基因(Cyt b)序列进行测序分析,所得基因序列在NCBI中比对,运用MEGA、DNASP等软件分析得到4个翘嘴鲌群体的遗传多样性数据。翘嘴鲌Cyt b基因序列长度为1088 bp,其中碱基A+T含量为55%,G+C为45%。共检测到258个多态位点,占核苷酸总数的23.71%。4个群体211个样本中得到46个单倍型,单倍型多样性为0.849~0.946,具体表现为TH>CD>CJ>DSH。核苷酸多样性为0.0083~0.174,具体表现为DSH> CD>CJ>DSH。4个群体间遗传距离0.052~0.152,太湖群体和淀山湖群体的群体间遗传距离最大为0.152。群体间的遗传分化指数范围为0.197~0.646,基因流值范围为0.152~1.019,其中长江与淀山湖群体的基因流值最小,长江与长荡湖群体的基因流值最大。基于NJ法构建的系统进化树结果显示,4个群体翘嘴鲌没有明显地理区分。本研究结果表明,长江下游翘嘴鲌群体遗传多样性较高,结合近年来翘嘴鲌渔业资源的调查结果,以深入了解其遗传背景和种质资源现状,为翘嘴鲌的野生资源保护提供基础数据。

Abstract:

In order to understand the genetic diversity of different Culter alburnus Basilewsky populations in the lower reaches of the Changjiang River, and evaluate the resource status of C. alburnus. A total of 211 C. alburnus samples from Dianshan Lake (DSH), Taihu Lake (TH), Changdang Lake (CD) and Changjiang River (CJ) were studied. Mitochondrial DNA b gene (Cyt b) of C. alburnus was sequenced, and compared with NCBI. Genetic diversity data of four C. alburnus populations were analyzed by MEGA, DNASP and other software. The Cyt b gene sequence was 1088 bp in length, with 55% A+T and 45% G+C. A total of 258 polymorphic sites were detected, accounting for 23.71% of the total nucleotides. 46 haplotypes were found in 211 samples from 4 populations, and the haplotype diversity ranged from 0.849 to 0.946, the order was as following: TH>CD>CJ>DSH. The nucleotide diversity was 0.0083-0.174, which was represented as DSH> CD>CJ>DSH. The genetic distance between the four populations was 0.052-0.152, and the maximum was 0.152 between Taihu population and Dianshan Lake population. The genetic differentiation indices ranged from 0.197 to 0.646, and the gene flow values ranged from 0.152 to 1.019, among which the gene flow values of Changjiang River and Dianshan Lake populations were the smallest, while those of Changjiang River and Changdang Lake populations were the largest. The results of phylogenetic tree based on NJ method showed that there was no obvious geographical differentiation among the four populations. The results of this study indicated that the population genetic diversity of C. alburnus in the lower reaches of the Changjiang River was high. Combined with the results of fishery resources investigation in recent years, the genetic background and germplasm resources of C. alburnus in the lower reaches of the Changjiang were further understood, which provided basic data for wild resource conservation.
