Identification the Effect of Endogenous Auxin in Maize Root Tip on Its Root Cap Specific Gene Using in Situ Hybridization

Abstract: The root tips of 3-d-old maize seedlings (about 1cm) are treated by antiauxin (clofibric acid 10-7 mol/L, 10-6 mol/L, 10-5 mol/L) one day, in situ hybridization is used to identify the influence of endogenous auxin in root tip on the expression of root cap specific gene (Clone C103). Experimental result shows that Clone C103 has a high expression in the periphery of central cap under normal condition; and its expression is inhibited efficiently by clofibric acid, especially clofibric acid 10-5 mol/L makes very weak expression of Clone C103 in the outmost of central cap. The primary result reveals that the normal expression of Clone C103 is profoundly regulated by endogenous auxin in maize root tip, and the expression of Clone C103 maybe has some relationship with statolith in the periphery of central cap.
