Bioinformatics Analysis of Berberine Bridge Enzyme in Papaver Somniferum L.

Abstract:

This research aimed to analyze the structure and characters of BBE nucleic acid and protein in Papaver Somniferum L. and construct the three-dimensional structure of BBE. We used tools of bioinformatics to analyze and predict BBE in the following aspects: biochemical function, hydrophilicity, signal peptide, subcellular localization, secondary and tertiary structure of protein and so on. At the time, construct the three-dimensional structure of BBE by comparative modeling using Discovery Studio, the model was refined by energy minimization, and assessment by Ramachandran Plot. The results showed that the full length of BBE gene and protein in Papaver somniferum L. was 5073 bp and 535 aa, respectively, BBE, which was located in the vacuole, and a stable acidic protein and rich in Gly, Ser, Leu contained a signal peptide of 23 amino acids in the N terminal. α-helix and random coil were primary secondary structural components of BBE. We also obtained a reasonable three-dimensional structure of BBE in Papaver somniferum L. by homology modeling. The research would helpful to improve the production of Berberine by genetic engineering means and develop new antibiotics.
