Research on Different Methods of Genomic DNA Extraction from Several Resource Plants in Huangshan

Abstract:

In order to protect our natural resources based on molecular biology and appreciate the comprehensive development and utilization value, we carried out a pilot study on molecular biology of several resource plants in Huangshan. The abstraction methods of genomic DNA from the leaves of several resource plants in Huangshan, such as Dendranthema morifolium (Ramat) Tzvel. cv. Gongju, Artemisia argyi, Mussaenda pubescens Ait. f., Chimonanthus salicifolius S.Y.H and Sinojackia xylocarpa Hu had been studied. The three methods, usual CTAB, modified CTAB and SDS were compared. Then using ultraviolet specrophotometer and 1% agarose gel electrophoresis tested the purity and integrality of the genomic DNA extraction. The experiment results indicated that, the modified CTAB method received better effect to extract the genomic DNA from Dendranthema morifolium (Ramat) Tzvel. cv. Gongju, Artemisia argyi and Mussaenda pubescens Ait. f. The usual CTAB method received better effect extract the genomic DNA from Chimomanthus salicifolius S.Y.H. For Sinojackia xylocarpa Hu, SDS method received better effect. The DNA distracted by the three methods showed better integrity by 1% agarose gel electrophoresis. So the DNA extraction could be used for molecular biology analysis.
