Preparation of Stained Specimens for the Adult of Pseudanoplocephala crawfordi and its Structural Observation

Abstract:

To reveal the characteristic structures of Pseudanoplocephala crawfordi, the stained specimens of the adult of P. crawfordi from pigs were prepared with Hematoxylin staining and Ziehl phenol-basic fuchsin staining. The results showed that the optimal staining condition for collum segment, immature and mature proglottids was in the 1:15 diluted hematoxylin dye solution at room temperature (RT) for 12 hours, and that of gravid proglottids was in Ziehl phenol-basic fuchsin dye solution at RT for 24 hours. Adults of the parasite was flat and white, the vertical axises of proglottids were shorter than the lateral ones. Testicles and sperm pocket began to show in the immature proglottids, and while the complete male and female organs of generation including testicles, sperm pocket, penis pocket, ovary, yelk gland, uterus and gonopore occurred in the mature ones. Gravid proglottids were filled with eggs, while without other organs of generation with the exception of a gonopore. These findings provided a important reference for control of porcine and human pseudanoplocephalasis and supplement the specimen library of parasitological research and teaching.
