Studies on the Tissue Culture and Mycorrhizal Technology of Ostryopsis davidiana

Abstract:

In order to get a number of plantlets and make mycorrhizal synthesis test of Ostryopsis davidiana, the Ostryopsis davidiana of which the stem segments with axillary buds were taken as the explants. The tissue culture from aspects of disposal, selection of basic medium, proliferation, rooting and mycorrhizal research of Ostryopsis davidiana were studied in the experiment. The results showed that: the best way of sterilization was soaked in 75% alcohol for 30 s, then treated in 5% NaClO for 7 min, then the germination rate could reached to 66%; The optimal basic medium was the WPM; The optimal proliferation medium for the buds was WPM+6-BA 0.5 mg/L+NAA 0.03 mg/L and the proliferation rate reached to 436.7%; For rooting, the optimal medium was WPM+NAA 0.2 mg/L. The rooting rate was up to 100.0%. Mycorrhizal synthesis test in the tissue culture bottle of Ostryopsis davidiana after rooting for 10 days showed that multiple growth indices of Ostryopsis davidiana plantlets were improved definitely after inoculated CgO5. The results provided the theoretical basis for the rapid propagation and mycorrhizal technology of Ostryopsis davidiana.
