Abstract:

Purification SOD in the blood of Sika Deer with in antler developing period and study on properties of SOD, were studied for developing anti-aging resources of deer blood and laid a theoretical basis. SOD has been purified from Sika Deer blood by chloroform-ethanol fractionation, thermal denaturalization, acetone precipitation, DEAE-32 chromatography and the SDS-PAGE was used to measure its subunit molecular weight. The results showed that SOD initial specific activity was 12.84 U/mg and 0.16 mg/mL-purified SOD was achieved. Finally, specific activity was 4122.74 U/mg ，and 321.2 times purification rate and 66.38 percent recovery rate were achieved. The relative molecular weight of subunit for SOD was 17.32 KD and 16.28 KD. The results demonstrated that the 60℃ and 0.002 mmol/L Cu2+ was the best condition for SOD purification.
