Abstract:
The objective of this paper is to reveal the genetic diversity of sisal cultivars. The genetic relationships of 8 sisal germplasm resources were analyzed by ISSR and RAPD. Polymorphism was identified by 8 ISSR primers and 8 RAPD primers in the germplasm. 53 bands were generated by ISSR with polymorphic bands accounting for 83.02% and similarity coefficient within 0.59 to 0.80. And 66 bands were generated by RAPD with polymorphic bands accounting for 92.42% and similarity coefficient from 0.52 to 0.76. Using cluster analysis (UPCMA) based on those polymorphic bands amplified with ISSR and RAPD primers, the relationships of 8 sisal species were constructed. The clustering results of RAPD and ISSR markers were similar, but had a little difference. The germplasm were divided into 2 clusters, and the significant correlation between ISSR and RAPD markers was observed (r=0.70). The results revealed that there was rich genetic diversity among resources of sisal.
