Abstract:
To establish a new technique for in vitro rapid propagation and conservation of superior germplasm of Gardenia jasminoides Ellis, new emerging stems of a ten-year old Gardenia were used as explants to develop an efficient protocol for tissue-culture propagation based on MS medium supplemented with different hormones, in addition to research into the transplanting technique. Results indicated that the proper original induction medium was MS+6-BA 1.5 mg/L+NAA 0.5 mg/L; Shoot reproduction medium was MS+6-BA 1.5 mg/L+IBA 0.3 mg/L with a propagation ratio of 4.6; The best rooting medium was 1/2MS+IBA 2.0 mg/L, and the rooting rate was 87%; The best growth in the matrix was based on the ratio of peat∶vermiculite =1:1(V:V).
