The Study of Nuclease Activated by Elicitors in Cell Suspension of Acer ginnala

Abstract:

The aim was to reveal the reason causing apoptosis of Acer ginnala cell co-cultured with a fungal elicitor. The microscope observation, genomic DNA electrophoresis and nuclease activity test was used to investigate the apoptosis of cell suspension. It was observed from the microscope that the shape and structure of the cells altered significantly. The genomic DNA of induced cells ran a ladder graph on the lane of gel electrophoresis and then smeared on the lane when the cells died. It was also detected that the nucleases of the cell were activated after induced. Specific bands with the size of 50, 40, 41, 38 and 34 kD were detected on the PAGE. The fungal elicitor activates nucleases to degrade the genomic DNA, and bring the cells to apoptosis eventually.
