In Vitro Selection for Salt Tolerance in Diploid Potatoes

Abstract: Potato (Solanum tuberosum L.) is considered moderately salt sensitive, therefore soil salinization seriously impacts the growth and development of potato, causing yield loss to potato production. In this research, 45 long day adapted S. phureja (PHU)-S. stenotomum (STN) hybrid clones were evaluated for their salt (NaCl) tolerance in vitro at 0, 40, 80, and 120 mmol/L using tetraploid variety‘Bintje’as salt tolerant control, and‘Mainechip’as sensitive control. Six growth parameters (shoot and root lengths, fresh and dry weights) were measured at the time of harvest, and six diploid potato clones (89-2-1, 188-1, 267-1, 566-1, 472-1 and 463-1) were found significant higher in salt tolerance than‘Bintje’and fore (354-1, 138-1, 507-1, and 270-2) lower than‘Mainechip’. These diploid PHU-STN clones selected may be valuable germplasm for breeding salt tolerant variety and studying the mechanism of salt tolerance in potatoes.
