Inheritance of Resistance to Ralstonia solanacearum in Flue-cured tobacco Oxford207 and Molecular Marker Screen

Abstract: Tobacco variety Oxford207 and Honghuadajingyuan(HD), which resistant to bacterial wilt(BW) and susceptible to BW respectively were used as parents. F1, F2, BC1 populations were constructed. The resistance of parents and populations were evaluated by temperature-controlled water inoculation Ralstonia solanacearum at seedling stage. The death rates of F1, F2 and BC1F1 are similar, which are steadily between resistant and susceptible parents’ regular surveys. The area under the disease curve of F1, F2 and BC1F1 are similar, which are steadily between resistant and susceptible parents’ also. Those data indicate that the resistance of Oxford207 to bacterial wilt is additive, not typical dominant and recessive. Simple sequence repeat (SSR) technology and bulked segregant analysis were applied to identify markers linked to the resistance gene. Among more than 800 pieces of SSR primers, 263 primers have polymorphism between resistant and susceptible parents and show co-dominant in F1, 3 primers have polymorphism between the resistant pool and susceptible pool of F2 individual plants.
