  A Review of Benefit of Ecological Economic Forest in Desertification Areas 2013, 29 (34):  1-6.  DOI: 10.11924/j.issn.1000-6850.2013-1673 Abstract + ( 767 )   PDF ( 636470KB )   ( 522 ) In order to help developing the ecological economic forest in Chinese desertification areas to benefit local ecologies and economics, the author reviewed recent 30 years research in the ecological economic forest. It had shown that: while the ecological economic forest had a lot of possible effect in reducing desert, maintaining the soil and water conservation, improving the soil, and recovering ecologies, it could earn a lot of profits. Therefore, it would be helpful for both ecologies and economics and was an effective way to restore ecological environment in China. However, there were still a lot of issues to be studied and more works needed to be done for the realistic applications. Therefore, in order to make the ecological economic forest to improve the environment and economy, and finally to benefit humanity, future works should focus on widely and intensively studies on the ecological economic forest, and bring the laboratory result into realistic as soon as possible. Based on the environment and effects of ecological economic forest in desertification areas, the author had made some advices in developing ecological economic forest, for the purpose of environment reconstruction in desertification areas.

Regenetated shoots was investigated with light microscopy method. The results were as follows: The origin of adventitious root was from the pith ray.The callus was irregular parenchyma with rich inclusions, and there were no root primordium in callus.

  Study on the Accumulation and Tolerance of 9 Plants to Heavy Metal Cd in Soil 2013, 29 (34):  12-16.  DOI: 10.11924/j.issn.1000-6850.2013-1565 Abstract + ( 996 )   PDF ( 532941KB )   ( 1090 ) Heavy metal pollution of soil has been increasingly serious environmental concerns. Phytoremediation technology is a kind of economy and environmental friendly treatment to remove heavy metals in soil with hyperaccumulation plants. To screen hyperaccumulator which can extract heavy metal cadmium from contaminated soils, a pot experiment was developed. The results showed that: the Cd contents of Sedum tatarinowii Maxim, Hibiscus syriacus L. and Brassica oleracea L. var. acephala DC. in above-ground parts were great than 10 mg/kg. The Cd-transfer ratio of Sedum tatarinowii Maxim was 3.06, the highest one among all tested materials. Cd in under-ground parts of Nerium indicum Mill., Amorpha fruticosa L. and Hibiscus syriacus L. could be great than 50 mg/kg. The results indicated that 5 plants mentioned above could be grow, and fix and eliminate Cd in Cd-polluted soils.

  Evaluation of Ecosystem Service Value for Long-term Plantations in Saline-alkali Soils of the Yellow River Delta 2013, 29 (34):  17-23.  DOI: 10.11924/j.issn.1000-6850.2013-1791 Abstract + ( 972 )   PDF ( 752778KB )   ( 517 ) In order to compare the ecological effects of different plantations for various tree species and probe deeply into the differences in ecological service value among plantations, taking the 27-year-old long-term plantations in saline-alkali soils of the Yellow River delta as objects, the forest ecosystem service values of different plantations were revealed by calculating a suite of specific indices using formulae and prices of the Specification for Assessment of Forest Ecosystem Services in China (LY 1721—2008). The results showed that: the largest value of ecosystem service was velvet ash (Fraxinus velutina) plantation and the second one was black locust (Robinia pseudoacacia), i.e., 55717.76 and 49865.21 yuan/(hm2?a), respectively. The ecosystem service values of plantations for different tree species varied much following the order: velvet ash>black locust>Siberian elm (Ulmus pumila)>tree-of-heaven (Ailanthus altissima). 3 mixed plantations of black locust and velvet ash, black locust and Siberian elm, black locust and tree-of-heaven had the similar service values, which were lower than the pure one of black locust. Among 5 categories of ecosystem service, the largest proportion of ecosystem service value was water conservation, and the second one was carbon fixation and oxygen release. However, both atmospheric environment purification and biodiversity protection had relatively lower service values.

  The Study of Rapid Multiplication and Water Saving Seedling Technology of Pinus sylvestris in Greenhouse in Minqin Sandy Land 2013, 29 (34):  24-29.  DOI: 10.11924/j.issn.1000-6850.2013-1886 Abstract + ( 964 )   PDF ( 677738KB )   ( 527 ) In order to show the technology of rapid multiplication and water saving seedling in greenhouse, the difference between field seedling and greenhouse seedling of Pinus sylvestris, the technology with rapid multiplication and water saving seedling are studied in Minqin sandy land. It showed that: greenhouse seedling possessed absolute advantage in seedling emergence rate and cultivation period, while the degree of influence from different season was relatively less, it could be seeded in every season, the average seedling emergence rate was over 82.56%. Greenhouse seedling provided relatively well condition for the growth of height and ground diameter than field seedling. The soil moisture content was higher than field seedling 4.79%, the water consumption was 119.51 mm in 127 days, the water consumption intensity was 0.94 mm/d, the water consumption accounts for about one-third of total water consumption in nearly half of the period, water consumption was 208.59 mm in 129 days, water consumption intensity was 1.62 mm/d. water consumption accounted for about two-third of total water consumption in nearly half of the period. The water resources were scarce in Minqin sandy land, improving the management level above seedling, cultivating high quality and high adaptability of nursery stock through studying the water consumption regular of Pinus sylvestris in greenhouse. It had great significance for appropriate use of limited water resources.

  Chemical Control of the Rubber Powdery Mildew in Rubber Nursery 2013, 29 (34):  30-33.  DOI: 10.11924/j.issn.1000-6850.2013-1719 Abstract + ( 840 )   PDF ( 526320KB )   ( 610 ) To screen effective therapeutic agent, 5 fungicides, namely, prochloraz (45% ME), flusilazole (40% EC), tridemorph (75% EC), triadimefon (15% WP) and sulfur (95% WP) controlling powdery mildew of Hevea rubber were conducted by using conventional method in rubber nursery. The preliminary results showed that: after application 7 days sulfur (95% WP) was the best with control effect 83%, had a significant difference compared with other 4 kinds of agents. Tridemorph (75% EC) was better with control effect 60.2%. Flusilazole (40% EC), triadimefon (15% WP) and prochloraz (45% ME) were worse with control effect less than 50%. After application 30 days, sulfur (95% WP) was still the best with control effect 84.1%. Tridemorph (75% EC) was better with control effect 78.2%. Control effect of Flusilazole (40% EC) was 69.1%. There was no significant difference between the 3 agents. Prochloraz (45% ME) was the worst with control effect 33.0%. So, sulfur (95% WP) was still a significant therapeutic agent against powdery mildew of Hevea rubbe in rubber nursery. Tridemorph (75% EC) and flusilazole (40% EC) could be further development and utilization.

Orchard grass in orchard soil and water conservation research of related factors, such as soil moisture, structure, fertility, geothermal orchard microclimate, soil and water loss, many aspects, such as comprehensive, grass varieties screening found orchard grass planting in soil and water conservation effect research still needed further development, especially for mature orchards effect of non-point source pollution control still existed deficiencies, the future still needed from ways of sod, grass species selection and application, orchard grass planting techniques, the influence of soil water, groundwater and water quality was studied further.

  The Analysis and Comparison of Climate Characteristics in Jiangxi Flue-cured Tobacco Production Regions 2013, 29 (34):  44-49.  DOI: 10.11924/j.issn.1000-6850.2013-0672 Abstract + ( 999 )   PDF ( 587305KB )   ( 1178 ) In order to study the climate characteristics of Jiangxi flue-cured tobacco production regions and their suitability to flue-cured tobacco production. From climate data of Jiangxi meteorological department, the author selected were data of 16 tobacco producing counties in recent 40 years (1970-2010). Focusing on average temperature, rainfall and sunshine hours, the author discussed the climate characteristics of Jiangxi tobacco producing regions; based on Jiangxi tobacco production season, compared the climate features of various counties during tobacco production season and the differences with those of other tobacco production regions in China and abroad. The assessment showed that: Jiangxi Province had a warm climate, yearly average temperature was 18.82℃, sunshine hour was 1623.11 h, rainfall was 1632.14 mm, was suitable for tobacco production. However, low temperature at the beginning of the production season, high temperature and too much rainfall during the maturing period might appear occasionally, which would be detrimental to tobacco growth and leaf quality. The average temperature of flue-cured tobacco growing season in Jiangxi Province was close to that of USA flue-cured production region, higher than that of Yuxi (Yunnan), Sanming (Fujian), Brazil and Zimbabwe, but Jiangxi Province had lowest temperature during seedling period, highest temperature during maturing period; the average sunshine hours of flue-cured tobacco growing season in Jiangxi Province was a bit more than those of Sanming (Fujian), but less than those of Brazil, Yuxi (Yunnan) and Zimbabwe, much less than those of USA flue-cured production region; the average rainfalls of flue-cured tobacco total field growth period and maturing period in Jiangxi Province were about the same as those of Sanming (Fujian), much higher than those of Brazil, Zimbabwe, USA and Yuxi (Yunnan). Jiangxi province had a warm climate, with enough sunshine, abundant rainfall and long frost-free period, and was suitable for tobacco production. Occasionally, there might appear low temperature and little sunshine at the beginning of the production season, high temperature and burning sunshine, or too much rainfall and severe flood during the maturing period. Comparing it with those of famous tobacco production regions in China and abroad, Jiangxi climate had its distinct features. Tobacco production practices should be carefully adopted to foster the strengths and circumvent weaknesses, preventative measures should be arranged to avoid the losses caused by abnormal climate conditions.

  Inheritance of Resistance to Ralstonia solanacearum in Flue-cured tobacco Oxford207 and Molecular Marker Screen 2013, 29 (34):  50-55.  DOI: 10.11924/j.issn.1000-6850.2013-0867 Abstract + ( 772 )   PDF ( 1836057KB )   ( 528 ) Tobacco variety Oxford207 and Honghuadajingyuan(HD), which resistant to bacterial wilt(BW) and susceptible to BW respectively were used as parents. F1, F2, BC1 populations were constructed. The resistance of parents and populations were evaluated by temperature-controlled water inoculation Ralstonia solanacearum at seedling stage. The death rates of F1, F2 and BC1F1 are similar, which are steadily between resistant and susceptible parents’ regular surveys. The area under the disease curve of F1, F2 and BC1F1 are similar, which are steadily between resistant and susceptible parents’ also. Those data indicate that the resistance of Oxford207 to bacterial wilt is additive, not typical dominant and recessive. Simple sequence repeat (SSR) technology and bulked segregant analysis were applied to identify markers linked to the resistance gene. Among more than 800 pieces of SSR primers, 263 primers have polymorphism between resistant and susceptible parents and show co-dominant in F1, 3 primers have polymorphism between the resistant pool and susceptible pool of F2 individual plants.

  Genetic Analysis of Resistant to Tobacco Bacterial Wilt 2013, 29 (34):  56-60.  DOI: 10.11924/j.issn.1000-6850.2013-0690 Abstract + ( 1086 )   PDF ( 506326KB )   ( 473 ) In order to provide evidence for resistance breeding scheme, the genetic of resistant to tobacco bacterial wilt were analyzed. The 15 tobacco germplasm including different resistant to bacterial wilt and its F1 cross by diallel mating design were identified in disease field of Anhui Province in China in 2010 and 2011. The results showed that: the resistance additive effect to tobacco bacterial wilt was dominant, the dominant effect was weak, additive×additive epistatic effect and G×E interaction effect was detected. The heritability in narrow sense of resistance was among 30.40%-57.14%.

  Genetic Analysis of Nicotine Conversion of Burley Tobacco Using Mixed Model of Major Gene and Polygene 2013, 29 (34):  61-65.  DOI: 10.11924/j.issn.1000-6850.2013-1484 Abstract + ( 906 )   PDF ( 556271KB )   ( 546 ) In order to understand the genetic control of nicotine conversion in burley tobacco the joint genetic analysis of nicotine conversion was carried out based on four generations P1 (Va1050 with high nicotine conversion), P2 (Va1053 with low nicotine conversion) and their F1 and F2 using mixed model of major gene and polygene. Genetic parameters were also estimated. The results showed that the genetic model of two pairs of major genes with equal additive and equal dominant effect and polygenes with additive and dominant effect (E-5) was selected for nicotine conversion of burley tobacco materials used in this study. The heritability of major genes and polygenes was 58.18% and 11.72%, respectively. As a consequence, nicotine conversion of burley tobacco mainly was controlled by two pairs of major genes and modified by polygenes and environmental factors such as temperature and humidity. In practical breeding, it will be effective that selecting parents with low conversion for breeding target varieties because of high heritability of this trait. Additionally, temperature and humidity control are also very important for reducing nicotine conversion during tobacco leaves curing and aging.

  The Dynamic Changes on Secondary Enzymes Activeness of Different Flue-cured Tobacco Leaves in Carbon and Nitrogen Metabolism 2013, 29 (34):  66-70.  DOI: 10.11924/j.issn.1000-6850.2013-1042 Abstract + ( 743 )   PDF ( 597038KB )   ( 409 ) Using Yunyan87,Zunyan6 and k326 as materials, the dynamic changes on secondary enzymes activeness of the three flue-cured tobacco leaves in carbon and nitrogen metabolism were studied. The result showed that, （1）The activity of amylase in all three flue-cured tobacco leaves rose as transplant time went by. After 30th day, the increasing speed of all flue-cured tobacco was faster than before. The activity of amylase in Yunyan87 was highest between 15 and 45 day after transplanting. （2）The Sucrose synthase activity in Zunyan6 was relatively stable in 45 days, in which Yunyan87cut down quickly, at30th day, the lowest, then kept. on the contrary, the Sucrose synthase activity in Yunyan87 went up rapidly. Between 30th and 45th day, which in k326 was lowest. （3）The Invertase activity in Yunyan87 went up slowly at the beginning, and then changing less. In k326, it increased and then decreased, at 15th day, the maximum. The Invertase activity in Zunyan6 did not change much. Between 30 and 45 day after transplanting, Zunyan6 was lower than the other two. （4）The nitrate reductase activity of the three flue-cured tobacco leaves was significantly different, Yunyan87 rose and then stable, but Zunyan6 changed a lot, while k326 went down continuously. At the 15th day, the nitrate reductase activity in Zunyan6 was highest, Yunyan87 and k326 t had no significant difference. At 45th day, Zunyan6 was higher than other, and k326 was lowest. （5）The Phenylalanine lyase activity in Yunyan87 took on an slowly increase and then decreased trend; Zunyan6 and k326 changed in the same trend, up-down-up, but Zunyan6 changed much more than k326. It showed that Yunyan87 had more abilities in secondary enzymes.

  Response of Leaf Photosynthetic Characteristics of Two Different Tolerant Tobacco Cultivars at Seedling Stage to Low Temperature Stress 2013, 29 (34):  71-75.  DOI: 10.11924/j.issn.1000-6850.2013-1207 Abstract + ( 800 )   PDF ( 646106KB )   ( 550 ) The changes in leaf photosynthetic characteristics of two different tolerant tobacco cultivars at seedling stage under low temperature stress and subsequent room temperature condition were investigated in the paper. The results showed that the seedling growth of the moderate tolerant tobacco cultivar K326 and the sensitive one Yunyan 97 was nearly inhibited completely at low temperature stress from 4℃ to 6℃, while the growth rate of K326 was significantly higher than that of Yunyan 97 under subsequent room temperature. The leaf photosynthesis at seedling was reduced under low temperature stress, and the inhibitory effects became worse with the elongation of stress treatment. The leaf photosynthetic indexes such as net photosynthetic rate (Pn), stomatal conductance (Gs), intercellular CO2 concentration (Ci), transpiration rate (Tr), water use efficiency (WUE), stomatal limitation (Ls), maximum photochemical efficiency of photosystem II (Fv/Fm), quantum yield of electron transport (ΦPSII), photochemical quenching co-efficient (qP), apparent electron transfer rate (ETR) and non-photochemical quenching co-efficient (NPQ) under low temperature for 6 days were significantly different with the untreated control. The leaf photosynthesis of tobacco seedlings was raised under subsequent room temperature condition, and the increasing range in K326 was much more than that of Yunyan 97. Although the leaf photosynthesis of the both two cultivars under subsequent room temperature for 9 days was not restored to the levels before low temperature stress, the leaf photosynthetic indexes in the moderate tolerant cultivar K326 were only a little lower than untreated seedlings, while these indexes in the sensitive cultivar Yunyan 97 were still significantly different with the untreated ones.

  Isolation, Prokaryotic Expression and Purification of ubc7 from Stigma of Ornamental Kale (Brassica oleracea var. acephala) 2013, 29 (34):  76-80.  DOI: 10.11924/j.issn.1000-6850.2013-1362 Abstract + ( 942 )   PDF ( 699993KB )   ( 683 ) In order to isolate the ubc7 cDNA (Boubc7) from stigma of ornamental kale (Brassica oleracea var. acephala) S13-b homozygotes and obtain the recombinant BoUbc7 fusion protein from E. coli cells. The total RNA was extracted from stigma of ornamental kale using CTAB method. The ubc7 gene was amplified by RT-PCR. The coding region of Boubc7 was inserted into the pET14b and E.coli BL21 (DE3) pLysS cell was transformed pET-14b-BoUbc7. The recombinant BoUbc7 fusion protein was induced by IPTG and purification using Ni2+-NTA resin. Results showed that, the Boubc7 cDNA contained an opening reading frame of 501 bp, encoding a 166 predicted amino acids residue with the predicted molecular mass of 18.7 kDa and the calculated pI of 5.35. SDS-PAGE results showed that the recombinant BoUbc7 fusion protein about 23 kDa was induced by IPTG and purification by affinity chromatography using Ni2+-NTA resin. The Boubc7 gene and recombinant BoUbc7 fusion protein has been obtained, which laid a solid foundation for investigate the biological function of BoUbc7.

  Resistance identification against Root-Knot Nematode Meloidogyne incognita of tomato cultivars 2013, 29 (34):  81-84.  DOI: 10.11924/j.issn.1000-6850.2013-1747 Abstract + ( 1004 )   PDF ( 803738KB )   ( 822 ) In order to clearly indicated the resistance of tomato cultivars against Root-not nematode, artificial inoculation method of potted tomato plants was used in the greenhouse. Resistant degree against the Root-knot nematode, Meloidogyne incognita, of 17 cultivars of tomato cultivated in Jiangsu Province and surrounding districts was identified respectively. The results showed that cultivar of Tianbao 316 was high resistant, cultivars of Israel A316 and Fenguo310 were moderately resistant, cultivars of Qinying 3, Bruneau and RuixingHuamei were general resistant, cultivars of Aomiduo9, Ouyaqi and American 903 were susceptible, the rest cultivars that Suhong2003, AmericanDahong, Hezuo903, TaiwanShengnv, Duocaishuiguo, Aoli, Huangguifei and HollandZishengguo were highly susceptible. These cultivars, which contains gene Mi, show high level of resistance against root-knot nematode. All cultivars of tomato were detected for the resistance gene Mi by PCR used special primers SCAR F and SCAR R. The results indicated that 3 cultivars of the tomatoes carried homozygous Mi and the gene sequence of the Tiaobao (KF015736)、Ruixinghuamei(KF015735) and Duocaishuiguo(KF015734) had 100% homology with Heinz 1706（DQ437766）.

  The Effects of Different Factors on Cucumber Tartronic Acid Content 2013, 29 (34):  85-89.  DOI: 10.11924/j.issn.1000-6850.2013-0732 Abstract + ( 759 )   PDF ( 531211KB )   ( 653 ) In order to breeding high tartronic acid content cucumber special varieties, three cucumber varieties with different tartronic acid content were chosen as the test materials. Using the L9(34) orthogonal test design to analyze the effects of four factors(different varieties, different sampling regions , different nitrogen concentration and different physiological periods) on tartronic acid content. The result showed that stems and leaves contain tartornic acid in spite of the fruits and the content is lower than fruits. When the concentration of nitrogen is in the normal level, the tartronic acid content is higher than in the lower and higher level. In addition, the tartronic acid in full bearing age is slightly higher than in smoke trailing period and in fruiting early stage. Furthermore, the combination of the highest cucumber tartronic acid content: cucumber varieties ‘C11-35’, sampling positions: fruit, nitrogen concentration: 0.014 mol/L, cucumber physiological period: full productive age.

  Study on Soil Physical-chemical Properties and Enzyme Activities in Apple Orchards with Different Planting Years 2013, 29 (34):  90-95.  DOI: 10.11924/j.issn.1000-6850.2013-0920 Abstract + ( 1088 )   PDF ( 647512KB )   ( 555 ) 【Objective】The objective of the experiment is to study the effect of apple orchards with different planting years on soil physical-chemical properties and activities of Soil eneyme.【Method】Soil samples were taken from apple orchards with different years of planting in Jixian,whose soil physical-chemical properties and activities of soil enzyme,and the correlativity between soil physical-chemical properties and activities of soil enzyme was analysed at the same time.【Result】With the time of planting increasing,the content of soil organic matter,available P,Cu,Zn,Mn and water-soluble salt firstly declined and then increased in different depths of soil layers(0-20cm,20-40cm,40-60cm)in apple orchards;the soil available K and total N had a firstly decreasing and then increasing trend in upper layer(0-20cm),which was as the same tendency as in middle layer(20-40cm);the soil available Fe firstly declined and then increased in upper layer (0-20cm).The activities of urease, invertase had a same trend with soil nutrients in different depths of soil layers,and the activity of catalase had a significant correlation with soil organic carbon, organic matter and available P.【Conclusion】With the time of planting increasing, soil nutrient content generally renders the phenomenon which decline in early growth stage and then rose in full bearing period, which was related to attention degree of the local fruit growers at different periods.The fruit growers should strengthen the fertilization management in early growth stage of fruit trees.Soil enzyme activity was greatly affected by soil physical-chemical properties, especially by available P, organic matter content and other factors.The activities of enzymes can be used as a means of evaluating soil biological activity and characterizing soil fertility levels in apple orchard.

  Estimation of Genomic C Value of Macadamia integrifolia spp. by Flow Cytometry 2013, 29 (34):  96-101.  DOI: 10.11924/j.issn.1000-6850.2013-1305 Abstract + ( 1031 )   PDF ( 963320KB )   ( 666 ) In order to establish the flow cytometry (FCM) method for determination of macadamia genomic C value, The tender leaves of macadamia cultivar ‘O.C’ was used as material, the adjusted OTTO and PI were used as isolation buffer and DNA fluorochrome, the Pisum sativum L. Ctirad and Zea mays L. CE-777 were used as DNA reference standards, the genomic C value was determined by FCM. The result showed that the DNA content of diploid macadamia spp. was 1.91488599 pg and the genomic C value was 0.957443 pg; the genome size of macadamia spp. was 1.872758×109 bp calculated by 1pgDNA=0.978×109 bp. The results determined by two different DNA reference standards have no significant difference. The study found that the FCM method was accurate and reliable, it provided a valuable reference for genomic C value determination of other species.

All of 22 characters which were researched, 19 quantitative characters had significant difference,3 qualitative characters were obviously different among all the varieties. So all of these characters can be considered as the cadidate testing traits.

  Analysis of POD Activity and Gene Expression Pattern in Banana Resistance to Fusarium Oxysporum f specialis (f. A peroxidase gene was identified by randomly sequencing from cDNA library obtained from banana roots. This gene was called MaPOD1 (GenBank accession number is KC478598). The cDNA sequence obtained by amplifying was consistent with plasmid sequence, indicated that this gene was full-length cDNA of banana POD gene, which contained an open reading frame of 948 bp encoding a protein of 328 amino acids. Through protein sequence homologous compare, it was found that the gene contained peroxidase active site and heme binding site structure. Semi quantitative RT-PCR demonstrated that the expression of MaPOD1 was higher in banana roots and pseudostems while lower in rhizomes. In resistant and susceptible varieties of banana after inoculation Foc TR4, the expression of MaPOD1 was upregulated, but in resistant varieties the time point of MaPOD1, compared to control, increased higher than that in susceptible varieties at time points, indicated that this gene played an important role in banana resistance. The expression of this gene had the same trend with enzymatic activity change, and gene expression lagged after enzyme activity change. MaPOD1 could be a new marker gene under the infection of Foc TR4.

  Studies on the Vitality of the Pollen of Lycoris chinensis 2013, 29 (34):  122-126.  DOI: 10.11924/j.issn.1000-6850.2013-1399 Abstract + ( 944 )   PDF ( 1772298KB )   ( 650 ) In order to understand the viability and the conditions of the pollen germination in vitro of Lycoris chinensis, the viability of fresh pollen of Lycoris chinensis were determined by TTC staining method and in vitro culture method. 99% pollen had vitality in 5% TTC solution at the temperature of 35 centigrade, pH 7.0. The single test of the three factors, concentrations of sucrose, boric acid and CaCl2 which effects the pollen germination and the pollen tube elongation were conducted. The most suitable concentration of them was 150 g/L, 60 mg/L and 50 mg/L. And the pollen germination in them was 36.7%, 77.6% and 44.2%, respectively. When the sucrose concentration was 150 mg/L, orthogonal designed that of boric acid and CaCl2. The results showed the most suitable concentrations for the pollen germination and the pollen tube elongation were the same as the single factor test, but the pollen germination rate was 87% and the pollen tube length was 897 μm, which was much higher than that. This suggested that there was a synergistic effect between sucrose, boric acid and CaCl2, which promote the pollen germination and the pollen tube elongation. All the data proved the pollen of L. chinensis had high vitality and offered the experimental base for the inter-species hybridization of L. chinensis and other species in Lycoris Herb.

  A Study on Edysterone Production of Ajuga lobata D. Don Cell Lines in Suspension Culture 2013, 29 (34):  127-133.  DOI: 10.11924/j.issn.1000-6850.2013-0856 Abstract + ( 991 )   PDF ( 1594106KB )   ( 566 ) In order to acquire the suspension cultivation production that cells growth quickly, biomass liveweight larger and edysterone content higher, the inducing condition for obtaining the granular embryonic callus was studied by chosen culture medium and screened the phytohormone, and the Ajuga lobata D. Don cell suspension culture system was established by optimizing from three aspects, which were the culture medium type, plant hormone species and their proportion, amount of inoculation in this paper. The results shown that: MS solid medium added 0.4 mg.L-1 2,4-D could induce the Ajuga lobata D.Don’s root segments to form unconsolidated granular embryogenic callus. Comparing the growth status of cell lines in three kind of liquid culture medium such as WPM, MS, 1/2 MS found that cells were grew best in MS. 2,4-D affected cell growth obviously. The cell biomass was 1.3 times higher than which cultured in culture medium added NAA. Experiments on the effect of phytohormone ratio on the Ajuga .lobata D. Don cell suspension culture shown that ,with 6-BA concentration increasing, ecdysterone content increased. Compared with that in control group, edysterone content in the cells cultured in medium added 0.5mg.L- 6–BA was significantly higher (P<0.05). The cells cultured in liquid medium added 1mg.L- 6-BA accumulated very significantly higher edysterone than that accumulated in the cells in control group (P<0.01). Though, the most prosper phytohormone proportion was 0.4 mg.L 1 2,4-D+6-0.5mg.L 1 BA，under which the net cell biomass could reach 4.3652±1.0739g after cultured 7 days, the ecdysterone content could reach 4.5692±0.2044mg/g of cell dry weight ,the cell lines was more uniform and grew faster. When the inoculation amount was 15%, cell lines grew best, at the 7th day after inoculation, cell growth rate was 1.109g/d,cell regeneration index can reach 3.45. It was the most proper inoculation quantity. Conclusion: the optimal cell suspension culture system was established , which would benefit the ecdysterone production.

After seedling transplanting, the plant samples were selected through a random sampling method to measure the growth rate of each organ about once a month. The results showed that: Maca rapidly grew from August to November. And the growth trends of the leaves’ fresh and dry weight were like an inverse ‘V’ , however, the trends of the roots’ fresh and dry weight were like a ‘S’. The leaves’ number and maximum length, as well as their fresh and dry weight, reached their maximum in the middle of November. And the roots’ length and diameter, as well as their fresh and dry weigh, reached their maximum in January of the following year. The ratios of both leaves’ fresh weight and dry weight according to whole weight tended to decrease with the growth stages advance, while that of the roots’ were opposite. There was a downtrend result could be observed for the proportion of fresh weight to dry weight of Maca, while the trend of ratio of root to shoot was upwards with the growth stages advance. When harvested in January of the following year, the yield of roots was the highest.

  Effects of ABA Pretreatment on the Seedlings of Sunflower Under the NaCl Stress 2013, 29 (34):  139-143.  DOI: 10.11924/j.issn.1000-6850.2013-0707 Abstract + ( 884 )   PDF ( 545077KB )   ( 619 ) Pretreatment in plants is recognized as a valuable strategy to eliminate the effects of salt stress. The study evaluated the effects of ABA pretreatment on the photosynthetic pigments, proline concentration, and the antioxidative mechanisms of sunflower under 150mmol?L-1 NaCl stress. It was found that the concentrations of chlorophyll a, chlorophyll b, carotenoid and total pigments were reduced by 7.8%、3.6%、0.5%、10.9%, respectively. All kinds of pigments concentrations were increased after using ABA pretreatment. Compared within four treatments, the most abundant pigmengts was from the seedlings of B1, while the least pigments was from the seedlings of A1. The concentration of proline was increased by 4.4 times (A1) and 4.28 (B1) times under salt stress, while the proline concentration of B0 treatment was increased by 2.23 times. The result showed that pretreatment with ABA stimulated the accumulation of proline under the control condition (A0). Analysis of the antioxidative enzymes CAT, POD, SOD revealed that ABA pretreatment affected antioxidant enzyme activities, especially POD activity. The most abundant activity was from the seedlings of B1, while the least activity was from the seedlings of A1. Compared with the control, the CAT activity was increased significantly (P<0.05), while the SOD activity was decreased significantly (P<0.05). The results suggested that ABA pretreatment enhanced the proline concentration, regulated the activities of antioxidants, and maintained the photosynthetic capacity of sunflower seedlings under the salt stress.

The results showed that the treatments of application sand 2 kilogram,3 kilogram,4 kilogram per plant separately on agronomic characters were better than that of application sand 1 kilogram and the control on the heavy clay soil. The yield, quality, average price and superior leaf ratio of flue-cured tobacco were increased significantly. The content of reducing sugar, total sugar and potassium were better by application sand 3 kilogram and 4 kilogram per plant separately than that of application sand 1 kilogram, 2 kilogram and the control, but the content of nicotine was decreased, but the effects of chemical composition by application sand1 kilogram,2 kilogram per plant separately were not marked.

  Effects of Nitrogen Application Rate and Remained Leaf Number on Yield and Main Quality Traits of Flue-cured Tobacco 2013, 29 (34):  149-153.  DOI: 10.11924/j.issn.1000-6850.2013-1034 Abstract + ( 788 )   PDF ( 560131KB )   ( 552 ) A field experiment was conducted to study the nitrogen application rate and remained leaf number on yield and main quality traits of flue-cured tobacco with HY-9-7 as material. The results showed that (1) the yield of flue-cured tobacco significantly increased with the increment of nitrogen rate, and it showed the increase-decrease trend with the increment of remained leaf number, and it showed 20 leaves>18 leaves >22 leaves. The output value of flue-cured tobacco increased and then decreased with the increment of nitrogen rate and remained leaf number. (2) As the increase of nitrogen application, total sugar and reducing sugar content decreased, while N, K, nicotine content of cured leaf increased. (3) As the increase of remained leaf number, content of total sugar, reducing sugar, K and nicotine were decreased. (4) The interaction effects of nitrogen rate and remained leaf number on yield, output value of flue-cured tobacco and content of total sugar and reducing sugar in cured leaves were significant.

  Effect of Transplanting Date on Meteorological Conditions and Characteristics of Flue - cured Tobacco 2013, 29 (34):  154-157.  DOI: 10.11924/j.issn.1000-6850.2013-0982 Abstract + ( 813 )   PDF ( 547559KB )   ( 422 ) In order to promote the high quality of flue - cured tobacco, and improve the economic efficiency of flue - cured tobacco, meteorological conditions in the growth stage of k326, agronomic and economic characters using 4 kinds of transplanting date were contrasted to get the best of transplanting date. The experimental results showed that, as the transplanting date delay, the average temperature during strong growth period descended and then rose, but the average temperature during mature and all growing period rose and then descended. The average temperature was all suited to tobacco growing. The precipitation during strong growth period showed a trend of increased significantly. If the transplanting date was premature, precipitation during strong growth period could not meet the high quality tobacco production. There is little difference in sunshine hours of 4 treatments, and it could meet the needs of high quality tobacco growth. Plant height, yield and value of flue - cured tobacco with the different transplanting dates were different significantly. Appropriately delaying transplanting date could get better agronomic traits and high economic efficiency.

  Study on Spectral Monitoring Model of Tobacco Water Content 2013, 29 (34):  158-161.  DOI: 10.11924/j.issn.1000-6850.2013-0677 Abstract + ( 709 )   PDF ( 644413KB )   ( 591 ) In order to study the change of water content of tobacco leaf with their drying time, we explored an effective V-NIR spectral monitoring model of the water content of tobacco leaf through drying experiment in this article. Contraposing the primitive spectral data to the preprocessed spectral data, we built a regression model through cross validation of partial least squares (PLS). It had been found that the PLS model of smoothing data cloud monitor the water content of tobacco leaf very well. In the regression model basing on all wavelengths, there were the regression coefficient r=0.9771, RMSE=0.0742 in training set, r=0.9573, RMSE=0.1009 in cross validation set, and r=0.9683, RMSE=0.0862 in prediction set. The experiment results showed that it was feasible to monitor the water content of tobacco leaf by the PLS model basing on the smoothing primitive spectral data of all wavelengths.

  Effects of Different Biochar-based Fertilizer on Nitrate content, Yield and Quality of Cabbage 2013, 29 (34):  162-165.  DOI: 10.11924/j.issn.1000-6850.2013-0935 Abstract + ( 1055 )   PDF ( 565575KB )   ( 657 ) Composting biochar and chemical fertilizer, field trial were carried out to compare the effect of various biochar-based composts on vegetable nitrate content and the differences of their fertilizer effect. There were four treatments including wheat straw derived (WBF), peanut shell derived (PBF) and cotton straw derived (CBF) biochar-based fertilizer ,the total nutrient is 37%,with N:P2O5:K2O=18:9:10 in this trial, and control experiment with the application of normal compound fertilizer, the total nutrient is 45%,with N:P2O5:K2O=15:15:15 was set to drive the comparisons. Compared to normal compound fertilizer, WBF decreased the nitrate content in cabbage by 35.38%, increased soluble sugar content by 13.33% and vitamin C by 31.55%. Meanwhile, the cabbage yield was increased by 45.03% with the application of WBC compared to compound fertilizer though the nutrient content in WBF was less than that in compound fertilizer by 18%. However, the effect of PBF and CBF were not obvious compared to compound fertilizer. Therefore, WBF is expected to become a new environment-friendly fertilizer to decrease nitrate content in vegetable and increase the quality and yield.

(‘Jinchun 5’, ‘Shijichunxiu’) as materials were used to study effects of different concentrations exogenous salicylic acid on germinating characteristics and seedling root growth under 150 mmol/L NaCl stress. The results showed that in 150 mmol/L under NaCl stress, cucumber seed germination and seedling root growth were inhibited under 150 mmol/L NaCl stress, inhibition of cucumber seedling root elongation was more obvious than the germ; exogenous SA could significantly improve the germination of cucumber seeds under NaCl stress, promote seedling root growth, alleviate on growth of cucumber germination and root damage of salt, and SA solution of 0.25 mmol/L treatment was the best for salt stress relieving effect. Under NaCl stress of salt tolerance in different varieties and exogenous SA alleviate the effect existent difference, ‘Jinchun 5’ salt tolerance and exogenous SA alleviate the effect than ‘Shijichunxiu’.

The results showed that Osmia bees began to fly in a low temperature (10-11℃) and had long working hours a day (10h). The mount of visiting flowers was high (4200) and pollination ability was high (A single pollination rate was 40%).The efficient pollination distance was 60m; The fruit setting rates of apple trees in the west and south were higher than that of apple trees in the east and north in the Osmia pollination orchards. The apples yield and qualities have been improved obviously, comparing with the natural pollination orchards. The Osmia bees liked to neat in the red tube, the nest ratio of bulrush and paper were higher than that of plastic tube, the nest ratio of bulrush was higher than that of paper tube.

The author took 20 growing places of Rosa roxburghii Tratt in Guizhou as samples, then investigated their distribution conditions, observed their phenophase, analyzed the degree of drought disaster and tested their qualities. According to the agro-climatic similarity theory, through the analysis of climate conditions on the quality of Rosa roxburghii Tratt, the results showed that: average temperature from May to August, average temperature in July, total precipitation from March to August, and the accumulated temperature≥10℃ in growth period had very important influence on Rosa roxburghii Tratt’s quality. The weight and diameter of Rosa roxburghii Tratt increased with the increase of total precipitation from March to August, precipitation too much or too low were detrimental to the synthesis of VC and SOD，average temperature from May to August and the accumulated temperature≥10℃ too high or too low were detrimental to the synthesis of water soluble total sugar and reducing sugar. According to suitable climate data of Rosa roxburghii Tratt, then the author drafted zoning map of eco-climatic suitability by 80% guarantee rate statistics. Divisions as follows: suitable areas were central Guizhou Province, most of Bijie District, southwest parts of Guizhou, north of Zunyi City; non-suitalbe areas were marginal areas of the northwest and southwest of Guizhou Province, part of southeast in Guizhou.

Based on the data of mean temperature, average minimum temperature and extreme minimum temperature in April from 1961 to 2010 in Guide County, Qinghai Province, the author analyzed the trends of mean temperature, average minimum temperature and extreme minimum temperature in April in the past 50 years. At the same time, by using the data of extreme minimum temperature in April, annual type division index of frost damage in pear florescence in Guide County was proposed: serious frost damage (Td≤-4.0℃), moderate frost damage (Td≤-2.0℃), light frost damage (Td≤0.0℃), and normal frost damage (Td≤2.0℃). On the basis of the yearly state classification, the author calculated transition probability matrix under various condition by using Markov Chain transition probability Model, then the Markov Chain Prediction Model for florescence frost damage in Guide County was established, and the prediction showed that serious frost damage would be a significant probability from 2011 to 2015. According to the test, the prediction of 2011 and 2013 was consistent with the actual situation.

  Effect of Hot-air Treatment on Quality and Chilling Injury 2013, 29 (34):  192-196.  DOI: 10.11924/j.issn.1000-6850.2013-0949 Abstract + ( 911 )   PDF ( 660110KB )   ( 687 ) Inorder to research the effect of hot-air treatment on quality and chilling injury of Loquat Fruits , Changes of chilling injury index, total soluble solids, titratable acidy contents, weight loss, bad fruit rate and firmness of Loquat Fruits were studied during storage at （6±1）℃, the relations between 46℃ hot-air treatment and protein constitution content were discussed during cold-storage and shelf-lifeas as well. The results showed that the fruits treated at 46℃ hot-air had maintained higher content of soluble and hot-stable protein, 46℃ hot-air treatment could decline total soluble solids, titratable acidy and rot rate, maintain higher firmness, weight loss and chilling injury index obviously， Loquat Fruits treated at 46℃ for 30min could receive good storage effect. All these proved that cold-resistance was close related to changes of protein constitution content and, hot-air treatment could also improve the storage quality of Loquat Fruits（eriobotrya japonica lindl.cv. Baiyu）.

The root weights , lateral root number, root length, root/shoot-ratio, and acid phosphatase activity(APA) were investigated in four strawberry genotypes(Hong Yan, Zhang Ji, Tian Cha Li,Tong Zi Yi Hao) grown under P-deficient and P-sufficient conditions in hydroponic culture. The results revealed that the root weights, lateral root number, root/shoot-ratio(R/S), and acid phosphatase activity(APA) of the seedlings were all obviously higher during the P-deficiency than they were in the P-sufficient. On the contrary, the root length decreased remarkably under the condition of P-deficiency compared with the P-sufficient. It indicates that the mechanism in morphology and physiology to increase P uptake from the outer environment by which the strawberry seedlings adapt to the P-deficiency. Furthermore, Tian Cha Li compared with other strawberry cultivars has the better environmental adaptability to low phosphorus stress.

It also influenced the yield and benefit. Based on conscientious observation and analysis, conducted a classification survey according to soil nutrient, moisture and texture and previous crops. Focusing on agronomic traits, soil nutrient and plant nutrient status. The result shows: There was a close relationship between the growth and development and characters performance of Erigeron breviscapus and soil fertility, moisture and texture and previous crops. These factors influenced not only soil nutrient content but also Erigeron breviscapus on nutrient absorption and the balance of each internal nutrient. Thus it effected the growth and development, yield and benefit of Erigeron breviscapus. Therefore, it will the best way for growing Erigeron breviscapus in loose soil, high fertility, good drainage field which planted flue-cured tobacco in the last season. It is the effective measures that ploughing deeply, loosing soil and strengthening management before transplanting in order to achieve improving the yield.

The results showed that: the PEG could retard the germination of stressed seeds, and the germination rate, germination index, and germination potential were all decreased with increasing PEG concentration, when it was under water potential≤-0.4 MPa. PEG 6000 impact seed germination with varying degrees of restrain about different safflower variety. Compared with varieties, ‘Yumin wuci’ had the best drought resistance, ‘Jihong No.1’ followed behind, ‘Xinhong No.4’ was weakest.

  Comparison on the content of volatile and semi-volatile acid in Flue-cured Tobacco from Different Production Regions 2013, 29 (34):  212-215.  DOI: 10.11924/j.issn.1000-6850.2013-0735 Abstract + ( 921 )   PDF ( 582048KB )   ( 408 ) C3F flue-cured tobaccos come from different production regions in Yunnan and Hunan were used for this study, the content of volatile and semi-volatile acid in flue-cured tobacco were studied by simultaneous distillation and extraction and gas chromatography. The results indicated that: (1) the content of volatile acid in Yunnan and Hunan exist greatly difference, but the content of semi-volatile acid differed little between the Yunnan and Hunan. Most content of volatile acid in Yunnan were all richer than those in Hunan, but the content of semi-volatile acid contrarily. (2) Most content of volatile acid in Qujing and Wenshan were significantly higher than those in Kunming and Zhaotong. The content of semi-volatile acid (except tetradecanoic acid) of Qujing were the highest, followed by Wenshan, while the Kunming were the lowest, and the difference was obviously. (3) Most content of volatile acid (except butyric acid) were significantly different among the different production regions in Hunan, but the difference had no obvious regularity. Most content of semi-volatile acid (except tetradecanoic acid) of Cili were the highest, while the Shaoyang were the lowest, and the difference was obviously.

  Investigate and ISSR Analysis of Pitaya Germplasm Resources in Fujian Province 2013, 29 (34):  216-220.  DOI: 10.11924/j.issn.1000-6850.2013-0980 Abstract + ( 874 )   PDF ( 761728KB )   ( 824 ) In order to know pitaya germplasm resources distribution and the genetic relationship in Fujian, an investigation on pitaya planting was conducted, and the samples were also collected. ISSR technology was applied to analyze the genetic diversity of 20 germplasm resources collected. 11 appropriate primers were selected from 40 random ISSR primers, 142 scorable amplification sites were detected, and the percentage of polymorphic ratio was 74.65%. The result of cluster analysis based on Jaccard revealed that the similarity coefficients (GS) between different cultivars ranged from 0.478 to 0.937, the average was 0.646. And when GS was 0.559, the whole population could be divided into 3 groups. This paper preliminarily revealed the pitaya germplasm resources distribution and the genetic relationship in Fujian and also proposed some varieties such as ‘Baijulong’, ‘Luobai’ and ‘Hongxianmi 1’ who suitable for popularization, which provided valuable reference for the species selection and breeding of pitaya.
