Studies on the Vitality of the Pollen of Lycoris chinensis

Abstract: In order to understand the viability and the conditions of the pollen germination in vitro of Lycoris chinensis, the viability of fresh pollen of Lycoris chinensis were determined by TTC staining method and in vitro culture method. 99% pollen had vitality in 5% TTC solution at the temperature of 35 centigrade, pH 7.0. The single test of the three factors, concentrations of sucrose, boric acid and CaCl2 which effects the pollen germination and the pollen tube elongation were conducted. The most suitable concentration of them was 150 g/L, 60 mg/L and 50 mg/L. And the pollen germination in them was 36.7%, 77.6% and 44.2%, respectively. When the sucrose concentration was 150 mg/L, orthogonal designed that of boric acid and CaCl2. The results showed the most suitable concentrations for the pollen germination and the pollen tube elongation were the same as the single factor test, but the pollen germination rate was 87% and the pollen tube length was 897 μm, which was much higher than that. This suggested that there was a synergistic effect between sucrose, boric acid and CaCl2, which promote the pollen germination and the pollen tube elongation. All the data proved the pollen of L. chinensis had high vitality and offered the experimental base for the inter-species hybridization of L. chinensis and other species in Lycoris Herb.
