The primarily study of the influence of the esophageal gland expressed gene 7E12 of Meloidogyne incognita in plant

Abstract: The south root-knot nematode (Meloidogyne incognita) was an important plant-parasitic nematode, the secretion proteins from its esophageal gland cells were the pathogenic factors which would hurt the host. Identification of the genes encoding these proteins was beneficial to know the pathogenicity and parasitism mechanism of the south root-knot nematode. The plant root specific expression vector pBI-Trp-7E12 of Meloidogyne incognita esophageal gland cell specific expression gene 7E12 and pBI-Trp were constructed by using the △0.3 truncated promoter of tobacco root specific expression gene TobRB7 to replace the CMV (cauliflower mosaic virus) 35S promoter of plant over-expression vector pBI121. In the research, pBI-Trp was used as blank control. These two vectors were transformed to the research model plant: Arabidopsis thaliana by using Agrobacterium rhizogenes via pollen tube channel infection method. Filtered for the transformated Arabidopsis thaliana by using the kanamycin resistance medium and obtained the T1 generation of transgenic plants. This study laid a foundation for the research and identification of the gene characterization of 7E12, so as to further the research on the pathogenic mechanism of south root-knot nematode to the host plants.
