Study on the Immobilization Conditions of Organic Phosphorus Pesticide Degrading Enzyme from Aspergillus niger J6

Abstract: In order to prepare remediation agent for degrading organophosphorus pesticides in polluted water or soil, immobilization conditions on the organic phosphoru pesticide degrading enzyme from Aspergillus niger J6 were studied. At first, crude enzyme liquid of organophosphorus pesticide degrading enzyme was prepared from Aspergillus niger J6, and then study on the immobilization conditions including the concentration of sodium alginate and CaCl2, and also the immobilizing time. The crude enzyme solution of the optimal entrapping condition could be gotten. The results showed that 2% sodium alginate, 4% CaCl2, the crude enzyme solution and sodium alginate solution ratio 20% (enzyme protein content is 0.15 mg/mL), the immobilizing time of 4 h, were the optimum immobilization conditions. Under the best condition, the ball was gotten more easily, better shape and elasticity, higher strength, and the relative enzyme activity reached the highest. Immobilization conditions of crude enzyme liquid from Aspergillus niger J6 were successfully studied, and the optimum embedding condition was obtained.
