Assay of Procyanidin in various tissues and organs of Gossypium hirsutum

Abstract: The aim was to study the contents of procyanidin (PA) in different tissue and organs of cotton. The contents of PA in various tissues and organs of G. hirsutum cv. CRI35, including calyx, petal, leaf, stem and root, were analyzed by visible spectrophotometer. The results revealed that the contents of soluble PA in calyx was the highest, reaching to 0.040μg/mg, while that in root was lowest, only 0.005μg/mg. The contents in leaf and petal were moderate, respectively 0.030 and 0.017μg/mg. And the contents of insoluble PA in various tissues and organs exhibited the similar content current as the soluble PA, which were respectively 2.324, 1.742, 1.449, 0.417 and 0.384μg/mg correspondingly in calyx, leave, petal, stem and root. Insoluble PA accounted for major part in CRI35, while soluble PA was only minor part. In petal, insoluble PA content was 84 folds of soluble PA, while in stem, the rates of two kinds of PA content was smallest, reaching to 29 folds. Taken together, results suggested that the difference of PA in various tissues and organs of CRI35 could be related to their executing functions, especially, which helped exploit the resistance/tolerance of cotton in breeding.
