The Establishment of High Efficient Regeneration System in Ningxia Rice D10

Abstract: The author choose the mature embryo of Ningxia high quality rice D10 as explants in tissue culture, in order to improving Ningxia fragrance rice with gene silence technology and established optimal and high efficient regeneration system. The effect of medium, exogenous plant hormone, genotype, light and temperature on vitro culture and regeneration system was studied by plant tissue culture method. The result showed that, different medium had different induction rate in D10. N6D medium was the highest induction rate, while MS medium was the lowest. The callus in 32℃ discontinued lighting culture condition were obviously better than 30℃ 12 h/d. 1 mg/L TDZ combining with low concentration IAA improved green shoot induction rate to 100%. Additionally, N6D medium pulsed 2 mg/L ABA could increased callus induction rate to 100%. The optimum condition and medium were 32℃ discontinued lighting culture in N6D medium pulsed 2 mg/L ABA, B5+TDZ 1 mg/L +IAA 0.2 mg/L differentiation medium and 1/2MS+KT 2 mg/L+NAA 0.2 mg/L+CH 2 g/L+Sorb 30 g/L shooting medium. The high efficient regeneration system in mature embryo of Ningxia high quality rice D10 was established.
