Optimization of Fermentation Conditions for Producing Protease by Immobilized Apergillus oryzae HDF-7

Abstract: In order to improve protease production and reduce fermentation period in the process of industrial production of bean paste, we adopt the method of the sodium alginate embedding method to immobilize Aspergilleas oryzae HDF-7, which was isolated from traditional fermented soybean as a starting strain and ascertain the best fermentation condition. Protease activity was as index, we determined that the most optimal inoculums size was 3%; the best proliferated time was 168 h; the most optimal fermented time was 24 h, it's protease activity was 18.49±0.65 U/mL. On the basis of immobilization, repeated batches of fermentation were 7 times. This study has improved the protease enzyme activity, shortened the fermentation time and increased the protease production by immobilized Apergillus oryzae HDF-7 and optimized immobilized conditions. It would settle base for the industrial production.
