Abstract: The seeds of Ziziphus jujuba Mill cv. Dongzao from Shanxi were collected as the materials. The seed morphological indexes, such as the transverse diameter and vertical diameter were measured, the thousand kernel weight (TKW) was weighted, and the rate of kernel was counted. Then the optimal condition for seed germination was studied and the appropriate medium for hypocotyl callus induction was selected. The results showed that: 1) the transverse, vertical diameter of the seed were (8.62±0.05)mm and(17.95±0.05)mm, respectively; TKW was (54.27±4.62)g; the rate of kernel was (64.17±3.82)%, so Dongzao was one of the high rate varieties of Ziziphus jujube. 2) the seeds on germination medium with GA3 0.4g/L for 24h pretreatment could reach the 100% germination rate; the liquid germination medium was a bit better than the solid, but the difference was not significant. 3) the best hypocotyl callus induction medium was 1/2MS+ TDZ 1.0mg/L+ sucrose 30g/L, and the induction rate was 60％.
