Rhizome Proliferation of Cymbidium sinense in Air Lift Bioreactor and Shoot Differentiation

Abstract: Rhizomes in vitro of Cymbidium. sinense were used as explants to investigate the effect of culture methods and light intensity on rhizomes of C. sinense in bioreactor, and effects of explant length and part on shoot differentiation. The results showed that immersion culture and light intensity of 1600 Lux of air-lift bioreactor was best for rhizome proliferation and growth. Shoot proliferation rate, shoot number and length were higher in the treatment of 1.0 cm length than others. Bottom part of rhizome explants was more favorable than other parts. The formation progress of green shoots was observed, shoot primordium formed on the surface of rhizome on the 10th, then the primordium never enlarged and development from the 20th.
