Effect of Successive Subculture Shoot Tips in Vitro of Lilium lancifolium on Cryopreservation

Abstract: In order to study the effect of subculture times of shoot tips on cryopreservation, the survival rates of 1-4 succesive subculture shoot tips in vitro of Lilium lancifolium after cryopreservation were compared. As the results showed, the survival rates were 55.8%, 93.3%, 67.0% and 64.1%. What’s more, the second generation subcultures which had been subcultured two months got the higher survival rate than other generations. However, continuous paraffin sections of shoot tips of every generation were investigated by microscopy, the results showed the second generation plantlets was found different from others in morphological studies of protomerism cell, which involved smaller cell volume, larger cell nuclear and richer cytoplasm, and this may lead to the higher survival rate of the second generation after cryopreservation than others .
