In vitro Culture and Plant Regeneration from Stem Segments of Dendrobenthamia angustata

Abstract: The study aims to establish an efficient tissue culture system for rapid propagation of Dendrobenthamia angustata. Using stem segments of Dendrobenthamia angustata as explants, effects of the basic culture medium, different types and the concentrations of plant growth regulators on the germination of axillary buds, proliferation of adventitious bud and rooting induction were examined. All nine hormone combinations promoted germination of axillary buds to some extent and the most suitable hormone combination was WPM + 1.0 mg/L 6- BA + 0.05 mg/L KT + 0.1 mg/L NAA, the germination rate achieved 86.29% at the aforesaid conditions. The combination of WPM+1.0 mg/L 6-BA+0.2 mg/L NAA was the best treatment for the culture of shoot induction, and the multiplication coefficient could reach 4.07. The treatment of WPM+1.0 mg/L IBA had the best effect on rooting induction, and its rooting rate was 78.94% with superior growth performance. The system of organization culture was suitable for regeneration of Dendrobenthamia angustata.
