Preliminary Study on Physiological and Biochemical Features of the Pathogen of Mango Bacterial Black Spot in Hainan Province

Abstract: In order to understand the physiological and biochemical features of Xanthomonas campestris pv. mangiferaeindicae and provide theoretical basis for scientific control of mango bacterial black spot, thirteen physiological and seven biochemical indexes of twelve Xanthomonas campestris pv. mangiferaeindicae strains from four cities (or counties) of Hainan Province were identified in this study. The results showed that nine out of thirteen physiological features were identical, including NaCl tolerance, pH tolerance, tween-80, acetate, sucrose, galactose, lactose, maltose and mannite, while the utilization of fructose, glucose, rhamnose and xylose was obviously different. Among seven tested biochemical features, oxidase, catalase, and amylase reaction, nitrate reduction, starch hydrolysis and gelatin hydrolysis were identical except H2S. Jaccard’s genetic similarity coefficients were calculated and dendrograms were constructed by the unweighted pair group method of arithmetic average (UPGMA) using the NTSYS-pc version 2.10. Different cluster analysis of physiological and biochemical tests indicated that strains from different cities or counties could share the same class, which showed that there were some diversities among Xanthomonas campestris pv. mangiferaeindicae in Hainan Province.
