Resistance Level and Mechanism of Colletotrichum gloeosporioides to Thiophanate-Methyl in Zhejiang

Abstract: The study is to observe the resistant frequency and the molecular mechanism of Colletotrichum gloeosporioides to thiophanate-methyl in Zhejiang. The sensitivity to thiophanate-methyl of 62 isolates of C. gloeosporioides was determined by the method of minimum inhibitory concentration (MIC). The β-tubulin gene region of 12 isolates was amplified. The results showed that 56 of 62 isolates (90.32%) were highly resistant to thiophanate- methyl, and 6 isolates (9.68% ) were sensitive. The β- tubulin sequences of 12 representative isolates of C. gloeosporioides were aligned. It showed that the resistant isolates to thiophanate-methyl produced mutation (A→C), and glutamic acid (GAC) was substituted by alanine (GCG) at amino acid 198. The point mutation of amino acid at codon 198 of C. gloeosporioides β-tubulin gene was related to thiophanate-methyl resistance.
