Germination and Rapid Propagation from Seed of Rhynchotechum obovatum (Griff.) Burtt

Abstract: The research aims to develop rapid propagation system from seed of Rhynchotechum obovatum (Griff.) Burtt via tissue culture. The seed of Rhynchotechum obovatum acting as explants were germinated and cultured on MS supplemented with different concentrations of hormone in order to select the optimum medium for different phases. The results showed that, the seeds and capsules were efficiently sterilized by 0.1% HgCl2 for 10 min. The optimum medium for germination and buds induction was MS 1.0 mg/L 6-BA 0.2 mg/L NAA. The suitable multiplication medium was MS 0.5 mg/L 6-BA 0.2 mg/L NAA and the suitable rooting medium was 1/2 MS 0.2 mg/L NAA. Using the methods obtained in this experiment, in vitro rapid propagation of Rhynchotechum obovatum could be achieved at the multiplication rate of 13.5 per 20 days, which could develop more seedlings in short time and provide guidance for the conservation and sustainable development of the resource.
