Analysis of flavonol glycosides in Folium Ginkgo by UPLC-UV

Abstract: In order to compare the contents of flavonol glycosides in the leaves of Ginkgo biloba L. from different species in the southwest of hubei province, the analysis of flavonol glycosides in Folium Ginkgo by UPLC-UV was studied. The separation was performed on a ZORBAX 2.1×50mm 1.8 Micron C-18 column with acetonitrile 0.1%( formic acid: water) (with gradient) as mobile phase at a gradient flow-rate of 0.3-0.35 mL.min-1 within 7 minutes, and the column temperature of 30℃, as the detector,Sthe DAD detection wavelength was 360nm,Squalitative analysis bySretention time and quantitativeSanalysis bySpeak area. The methodSseparated three kinds ofSflavonoid glycoside within 5 minutes. The calibration curve was linear in the range of 1-12μg.mL-1, with R2 being 0.9999 and RSD being less more than 2.5%. This method is easy, rapid, accurate and suitable for the quantitative analysis of flavonol glycosides. Among the different species, the maximum contents of flavonol glycosides were found in the EnG2 species. All varieties of quercetin levels were low while isorhamnetin occupy a higher proportion.
