A Preliminary Study on Sophora japonica Pollen Preservation Method

Abstract: The study aims at early acquisition and preservation of Sophora japonica pollen, avoiding inconvenience of artificial pollination and cross pollination, and prolonging the pollen viability during pollen pollination. Simple methods to evaluate the viability and to find out the time of pollen germination were established. Pollen viability was assessed by improving a growing media and evaluating three common pollen germination media components: agar, sucrose, and boric acid. Meanwhile, the pollen in room temperature (about 27℃ ) and in the refrigerator freezer (4℃ ) were differentiated to determine their longevity. Results showed that optimal germination of pollen was obtained at 1 g agar+1 g sucrose+100 mg/L boric acid. The time of pollen germination was 60 minutes. Under the same number of preservation days, the pollen germination rate in refrigerator freezer was significantly higher than that in room temperature. The pollen stored 5 days in refrigerator freezer was suitable for pollen pollination of Sophora japonica, and the pollen germination rate was greater than 50 percent. The suitable preservation method for field pollination of Sophora japonica pollen has been obtained, which can provide a reference for research on hybridization breeding of Sophora plant.
