Detection of Bombyx mori nucleopolyhedrovirus from Rearing Soil Using PCR

Abstract: Bombyx mori nucleopolyhedrovirus disease is the most serious disease among silkworm virus diseases. In order to prevent and control this disease, using soil representing as environment factor, feasibility of detecting pathogen BmNPV in the environment by PCR was discussed. High conserved gene of polyhedrin of BmNPV was used to design specific primers and BmNPV in soil could be detected successfully by PCR. No specific amplification was found from the other three lepidopteran-specific nucleopolyhedrovirus in soil by PCR, indicating that this method was highly specific. Sensitivity test showed that viral polyhedra number was 8.3×105 in the soil detected by PCR, and was 100-fold of that detected by microscopy.
