Bioinformatics Analysis of AfUGPase in Amorpha fruticosa L.

Abstract: The aims were to investigate the structure and function of AfUGPase gene in Amorpha fruticosa L., and find out the difference of inoculated plants and non-inoculated plants under the stress of AMF. AfUGPase gene was predicted by the tools of bioinformatics (nucleic acid sequence alignment, ORF analysis, multiple sequence alignment and evolutionary tree analysis) in the following aspects: the composition of amino acid sequence, structure of protein, molecular phylogenetic evolution and so on. The AfUGPase gene was 1831 bp, containing a 1416 bp open reading frame, which encoded 471 amino acid, while the estimated molecular weight and isoelectric point of the putative protein were 51584.2 Da and 6.07. It was a stable protein without memebrane- spanning domain, alpha helix and random coil were major structural elements of polypeptide chain, subcellular localization result indicated that it was located in cytoplasm, phylogenetic analysis showed that it had the secondary differentiation group with soybean UGPase. The results showed that AfUGPase gene contained a conservative Lys residue, participated in catalysis of enzyme, and was a typical UGPase protein, and also played an important role in plant metabolism and lipid synthesis.
