Establishment of Highly Efficient Plant Regeneration System of Populous suaveolens

Abstract: The study aims to exploit the antifreeze gene of Populous suaveolens and improve the resistance of poplar by biotechnology. Leaves of Populous suaveolens were used as explants to study the effect of different prescriptions of hormone on induction, differentiation of adventitious bud and rooting, and establish a plant regeneration system in vitro. The results showed that the optimum medium for adventitious bud regeneration was MS BA 0.5 mg/L NAA 0.05 mg/L with the frequency of adventitious bud regeneration of 98.8% and the number of shoots of 23.0. The optimum medium for rooting of shoots was 1/2MS IBA 0.3 mg/L with a rooting efficiency of 100%. The result of this study could lay a foundation for the improvement of the cold resistance of poplar and its molecular breeding.
