Rapid and Reliable Purity Identification of‘Suyu 30’Hybrids Using SSR Markers

Abstract: In order to quickly identify genetic purity of the high yield and good quality maize hybrid ‘Suyu 30’, ‘Suyu30’, ‘YJ7’, ‘HL40’ (the parents of ‘Suyu30’), ‘Suyu10’, ‘Suyu22’, ‘Suyu29’, ‘Suyu32’, ‘Zhengdan958’ and ‘Denghai9’ were subjected to genetic analysis with 40 SSR markers. The result suggested that five SSR markers of phi053k2, bnlg2291k4, bnlg2305k4, bnlg161k8 and umc2084w2 with stably amplified band and significant polymorphisms were determined as candidates for purity assessment of ‘Suyu30’, which could detect polymorphism between the parents. Besides, the single SSR marker (bnlg2291k4 or bnlg2305k) or the combinations of two pairs of SSRs (phi053k2 and bnlg161k8, umc2084w2 and bnlg161k8, phi053k2 and umc2084w2) could distinguish the maize hybrid ‘Suyu10’, ‘Suyu22’, ‘Suyu29’, ‘Suyu32’, ‘Zhengdan958’ and ‘Denghai9’ from ‘Suyu30’ as well. The purity test for ‘Suyu30’ using the five specific SSR markers was in agreement with the results of contrived fake seeds intermingling and field identification. Therefore, using SSR to identify the genetic purity of maize hybrid provides a reference for detecting the genetic purity of ‘Suyu30’, with the advantages of fast, accurate and practical characteristics.
