Determination of Gamma-aminobutyric Acid Content in Germinated Brown Rice by Ultra-high Performance Liquid Chromatography

Abstract: To provide an efficient method for the determination of gamma-aminobutyric acid (GABA) content in germinated brown rice, the content of GABA was determined by using ultra-high chromatographic column Waters BEH C18 (1.7 μm, 100 mm×2.1 mm) with 60% ethanol as the extraction agent, acetonitrile and water as mobile phase and OPA as the derivate agent. The results showed that the best derivation reaction time was 2 minutes, the sample could be stable for at least 24 hours under indoor temperature or stored at 4℃. The standard curve of GABA was plotted under the optimum condition. The mass concentration and the peak area of the low concentration standard sample showed good linear relationship, the detection limit was 0.5 mg/L, and the recovery rate was 98.88%-99.2% with the relative standard deviation of peak area being 0.6%-2.2%. The method is accurate, sensitive and time-saving, and can provide an effective and reliable tool for the determination of GABA in brown rice.
