A Modified Technique of Paraffin Section for Feijoa Flower Buds

Abstract: In order to obtain the optimal condition for observing Feijoa flower bud formation and differentiation by paraffin section method, flower buds in three different developmental periods were chosen in this study. Based on the conventional paraffin section method, the ways of fixation, softening, dehydration, clearing, embedding and dyeing, etc. were improved to adapt to the characteristics of the Feijoa flower bud. The results showed that after the materials were softened in hydrochloric acid solution for 24 h, dehydration, transparent and other steps were conducted for section operation, and a continuous and complete wax tape could be obtained. The final sliced section was observed through the microscope, finding that the internal structure of the visible material was complete and the dyeing was clear. The optimized paraffin section technique could avoid the problems of incomplete embedding, hardened and brittle organization, discontinuous wax tape and incomplete internal structure, etc. This study provided a set of operation steps by using paraffin section method to observe Feijoa flower bud formation and differentiation, which could be a good reference for other trees with similar flower structure.
