Cloning and Expression Analysis of 4CL Gene Promoter of Betula platyphylla

Abstract: In order to investigate the tissue-specific expression of 4-coumarate:coenzyme A ligase (4CL) gene in monomer lignin synthesis pathway of birch we cloned the promoter of 4CL gene using the Chromosome Walking method and used it to replace the 35S promoter of pBI121 plant expression vector directionally to construct the recombination vector P4CL:: GUS. The recombination vectors were transformed into the birch using the transient transformation system and the GUS staining were performed. The results suggested that we got the 1344bp promoter sequence upstream from the initiation codon. In this promoter region, in addition to the TATA-box, CAAT-box elements which is the basic components for the transcription initiation, there are a number of cis-acting element sequence including: the 35S promoter element ASF; cis-acting element ABRE participating in the abscisic acid response; cis-regulatory elements CGTCA-motif involved in methyl jasmonate response; and the light responsive element G-Box, ACE, 4CL-CMA2b,etc. The result of GUS staining showed that the stem segments of transgene birch were blue. All of these results suggested that the 4CL promoter has the promoter activity and it may be involved in the development of birch xylem.
